Antibacterial Compound Preparation and Dosing

Minocycline hydrochloride and Tetracycline hydrochloride (Sigma-Aldrich) were resuspended in water at 4 mg/ml, before dilution in RPMI to the required concentration. Tigecycline hydrate (Sigma-Aldrich) was resuspended in DMSO at 5 mg/ml, before dilution in RPMI. Stock solutions were made fresh for each experiment. Typically for 96-well plate assays, 50 μl of stock solution or cRPMI control was added per well.

Tetracycline Antibiotic Formulations and Dosing

Ethanol (95% v/v, USP) was diluted to 20% (v/v) with rodent drinking water from the Laboratory Animal Resources Center, Texas Tech University Health Sciences Center, Lubbock TX. Tetracycline derivatives and their properties are listed in Table 1. All tetracyclines used in our study have been approved by the FDA as antibiotics in doses varying up to 500 mg, although some are no longer used (see: http://www.accessdata.fda.gov/scripts/cder/drugsatfda/). Drugs were obtained from commercial sources and stored according to the vendor's recommendations. Minocycline HCl (#M9511), chlortetracycline HCl (#46133-Fluka), oxytetracycline HCl (# D9811), tigecycline hydrate (#PZ0021), and tetracycline HCl (#T3383) were obtained from Sigma-Aldrich (St. Louis, MO, USA). Doxycycline HCl (#SC337691) and demeclocycline HCl (#SC204710A) were obtained from Santa Cruz Biotechnology (Dallas, TX, USA) and tigecycline (base) (#15026) was obtained from Cayman Chemical (Ann Arbor, MI, USA). Doses of 0, 20, 40, 60, 80, 100 and 120 mg/kg were made fresh and used within 25 hours.

Quantitative Determination of Tetracycline Analogues

Tigecycline hydrate (CAS 220620-09-7), CTC hydrochloride (CAS 64-72-2), TET hydrochloride (CAS 64-75-5), demeclocycline hydrochloride (CAS 64-73-3) as the internal standard (IS) for CTC, formic acid, acetonitrile, 1,2-dichloroethane, and water were purchased from Sigma-Aldrich (St. Louis, MO). Minocycline hydrochloride (CAS 13614-98-7) was supplied by Thermo Fisher Scientific (Waltham, MA). Tigecycline-d9 (CAS unlabeled) as the IS for TIG and MIN, and doxycycline-d3 hyclate (CAS unlabeled) as the IS for TET were purchased from Toronto Research Chemicals (North York, ON, Canada). For chromatographic analysis, the working solutions of each drug were prepared by diluting stock solutions (in methanol) in 0.1% formic acid in water.
Analytical standards of CTC, TET, and MIN dissolved in deionized water were also used for administration to animals during the PK experiment. Only TIG was not of an analytical standard due to the unavailability of TIG hydrochloride salt; therefore commercial product Tygacil (Pfizer, New York, NY) which contains a hydrochloric acid was used (TIG, lactose monohydrate, hydrochloric acid, sodium hydroxide).

Bacterial Growth in LB Broth and Agar

The bacterial strains used in this study are listed in Table 1. Bacterial growth was performed using Luria-Bertani (LB) broth (BD BioSciences, San Jose, CA, USA) with or without 1.5% of BactoTM Agar (BD BioSciences, San Jose, CA, USA) (named LB-agar) and 37 °C was set as a culture temperature unless indicated otherwise. Tigecycline hydrate and other chemicals used in this study were purchased from Sigma-Aldrich (Sigma-Aldrich, Saint Louis, MO, USA).

Purification and Characterization of Bioactive Compounds

Ciprofloxacin hydrochloride monohydrate was purchased from FUJIFILM Wako Pure Chemical Corporation.
