Grazoprevir

Grazoprevir was purchased from MedChem Express (MK-5172, product number HY-15298). Asunaprevir (BMS-650032, product number A3195) and danoprevir (RG7227, product number A4024) were purchased from ApexBio.

Asunaprevir, boceprevir, danoprevir, MK-5172 (a.k.a., grazoprevir), and simeprevir were from MedChemExpress. Telaprevir was from Selleck Chemicals. BILN-2061 was a gift from Roger Tsien and Stephen Adams (UC San Diego). Concentrated NS3 inhibitor stocks were dissolved in DMSO at concentrations between 3 – 10 mM and diluted into cell culture media at the indicated working concentrations.

All DNA transfections were carried out using Lipofectamine 3000 (Thermo Fisher) according to the manufacturer’s instructions. All transcriptional effector experiments were carried out at a mass ratio between 1:2 and 1:3 of Gal4_DB constructs to the transcriptional effector. For imaging experiments, fibronectin (Product #F1141, Sigma-Aldrich) was seeded on to glass coverslip substrate at a dilution of 5 μg/mL for 1 h at RT and rinsed three times with PBS before seeding cells. All other experiments were carried out on tissue cultured treated plastic (Corning) or for cell adherence by the manufacturer (ibiTreat by Ibidi).
The drug was either added at the time of transfection or 24 h later. Grazoprevir, danoprevir, telaprevir, and glecaprevir were purchased from MedChemExpress. Stocks were prepared at between 3 −10 mM concentrations between 3 and 10 mM by dissolution in DMSO.
