Synthesis of Carbon Nanomaterials from Nanodiamonds

CNOs were obtained according to the method proposed by Kuznetsov,72 (link) in which diamond nanoparticles (NDs, Carbodeon µDiamond Molto) with a diameter of 4.2 ± 0.5 nm and a ND content of ≥97 wt% were annealed. All the chemicals and solvents were commercially available and used without further purification: nanodiamond powder (µDiamond®Molto, Carbodeon); 1,2-dicyanobenzene (∼98%, Sigma-Aldrich); ammonia solution (25% pure p.a., Sigma-Aldrich); maleimide (∼99%, Sigma-Aldrich); tetrahydrofuran (pure p.a., Avantor); 2-propanol (pure p.a., Avantor); acetone (99.5% pure p.a., Avantor); 2-mercapto-4-methyl-5-thiazoleacetic acid (∼98%, Sigma-Aldrich); zinc chloride (≥99%, Sigma-Aldrich); nickel(ii) chloride (∼98%, Sigma-Aldrich); copper(ii) chloride (∼97%, Sigma-Aldrich); hydrochloric acid (35–38% pure p.a., Avantor); sulphuric acid (95% pure p.a., Avantor); sodium chloride (pure p.a., Avantor); aluminium powder (∼99.99%, Avantor); sodium (Sigma); 1-octanol (≥95%, Sigma-Aldrich); potassium permanganate (Avantor); dimethyl sulphoxide (DMSO, pure p.a., Avantor); and methanol (99.5% pure p.a., Avantor).

Potato Starch Modification Protocol

Extraction and Identification of Bioactive Benzofurans

The fermentation medium was prepared from chemicals obtained from Merck (Darmstadt, Germany). Standards: 2-arylbenzofurans mulberrofuran Y (1), moracin C (2), and mulberrofuran G (3) were obtained from ChemFaces (WuHan, China) in 98% purity (Figure 1). The organic solution, ethyl acetate, and methanol were purchased from the VWR International (Stribrna Skalice, Czech Republic) in the analytical grade. Dimethylsulphoxide was obtained from VWR Chemicals. Formic acid was obtained from Fisher Scientific (Merelbeke, Belgium) in >98% purity. Ultra-pure water was obtained from a Millipore system (Bedford, MA, USA).

Pharmacokinetic Study of PH46A with Compound 1 as Internal Standard

PH46A [purity 97.1 %; correction factor (purity and salt content): 0.6429] and Compound 1 as Internal Standard (IS, Fig. 1) (purity 99.8 %) were obtained from Trino Therapeutics Ltd (Ireland). Control rat (Sprague Dawley) and dog (Beagle) plasma and blood were obtained from Charles River Laboratories (UK) using lithium heparin as anticoagulant and stored at -20 °C when not in use (all control plasma was mixed and centrifuged prior to use). HPLC grade solvents and additives, including methanol (MeOH), acetonitrile (ACN), water, ammonia solution, acetic acid (AA), acetone, dimethylsulphoxide (DMSO), chloroform and formic acid (FA), were purchased commercially between VWR (Ireland) and Sigma-Aldrich (Ireland) and were used without further purification. Size 12 gelatine capsules (Torpac ‘Loc Ring Capsules’, USP grade) were used to administer the dose for the pharmacokinetic study.
