Ultra low attachment 96 well culture plate

Manufactured by Corning

The Ultra-low attachment 96-well culture plate is a cell culture product designed to promote the formation of three-dimensional cell aggregates, or spheroids. The plate's surface is treated to minimize cell attachment, enabling the cells to grow in a more natural, three-dimensional environment. This product is intended for use in applications where the generation and maintenance of spheroid cultures are required, such as in cancer research, stem cell studies, and drug screening.

CD14+ Cell Isolation and Culture

Check if the same lab product or an alternative is used in the 5 most similar protocols
Following removal of dead cells, CD14+ cells were isolated using positive magnetic bead selection with the CD14 MicroBeads (Miltenyi Biotec) according to the manufacturer's instructions. After two rounds of positive selection, purity of the CD14+ cell population was higher than 90% (Rodriguez-Garcia et al, submitted). Freshly isolated CD14+ cells were plated at a density of 1×105 cells per well in ultra-low attachment 96-well culture plate (Corning, Corning, NY) in 0.2 ml of immune cell media consisting of X-VIVO 15 Media (Lonza, Walkersville,MD) supplemented with 10% charcoal stripped human AB serum (Valley Biomedical, Winchester, VA) prior to treatment.

Hormone Treatments of Cell Suspension

Check if the same lab product or an alternative is used in the 5 most similar protocols
17β-estradiol (E2; Calbiochem, Gibbstown, NJ) and progesterone (P; Calbiochem) was dissolved in 100% ethanol at an initial concentration of 1×10−3 M, evaporated to dryness in a glass vial and resuspended in complete media consisting of X-VIVO 15 Media (Lonza, Walkersville, MD) supplemented with 10% charcoal stripped human AB serum (Valley Biomedical, Winchester, VA) to a concentration of 1×10−5 M. Further dilutions were made to achieve final working concentration of 5×10−8 M E2 and/or 1×10−7 M P. Both hormone concentrations are routinely used by our laboratory and are within the physiological range of hormone concentration [24 (link)]. As a control, an equivalent amount of ethanol without dissolved hormone was initially evaporated. Freshly isolated mixed cell suspension were plated at a density of 1×105 cells per well in ultra-low attachment 96-well culture plate (Corning, Corning, NY) in 0.2ml of complete media and treated either with 5×10−8 M E2 and/or 1×10−7 M P at 37°C for 24hr.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
