Thyroid Cell Lines and Hydrogen Sulfide Treatment

Human normal thyroid cell line Nthy-ori3-1 and human thyroid carcinoma cell lines TPC-1, TT, and ARO were obtained from CoBioer Biosciences Co. Ltd. (Nanjing, Jiangsu, China). Cells were cultured in RPMI 1640 medium supplemented with 10% fetal bovine serum, 100 units/ml penicillin, and 100 μg/ml streptomycin. Cells were grown at 37°C in a humidified atmosphere of 95% air and 5% CO2 [29 (link)]. Confluent cells were starved overnight in a serum-free RPMI 1640 medium. The cells were then treated with 10, 25, 50, 100, and 200 μM NaHS (an H2S donor, Sigma-Aldrich, St. Louis, MO, USA) for 24 h. The phosphate-buffered saline (PBS) group was served as a control.

Culturing Human Thyroid Cell Lines

The human thyroid carcinoma cell lines, CAL-62 and SW579, were obtained from The Cell Bank of Type Culture Collection of The Chinese Academy of Sciences. The immortalized normal human thyroid follicular epithelial cell line, Nthy-ori3-1, was purchased from CoBioer Biosciences Co., Ltd. (cat. no. CBP61205). HUVECs were purchased from Shanghai Zhong Qiao Xin Zhou Biotechnology Co., Ltd. HUVECs, CAL-62 and SW579 cells were cultured in DMEM supplemented with 1% penicillin/streptomycin and 10% FBS, at 37°C (5% CO2) in a humidified atmosphere. Nthy-ori3-1 cells were maintained in RPMI-1640 with 10% FBS and 1% penicillin/streptomycin.
