Hepatic Microsomal Metabolic Profiling

The reaction mixture consisted of human liver microsomes (BD Gentest) in 100 mM potassium phosphate buffer (pH 7.4) and 10 μM test chemicals. After preincubation at 37 °C for 5 min, the reaction was initiated by adding NADPH regenerating solution (BD Biosciences). Samples (50 μl) were collected at 0 and 30 min. The reaction was terminated by adding 450 μl of ice-cold acetonitrile with imipramine (100 ng/ml, internal standard). After vortexing and centrifugation at 4 °C for 5 min at 13,000 rpm, the clear supernatant was collected, transferred to liquid chromatography (LC) vials, and analyzed by LC-MS/MS (Agilent 6460) for the quantification of the chemicals.
