Particle Size Analysis of Tolbutamide and Tenofovir Alafenamide

Two drugs were assessed for particle size: tolbutamide (TBM) (Sigma Aldrich) and tenofovir alafenamide (Gilead Sciences, Inc.) with urocanic acid (Fisher Scientific)[31 ] (TAF-UA). Approximately 20 mg of each drug was individually allocated in two vials. Two (2) mL of PBS was added to one vial, and 2 mL of PBS 0.05% Tween 20 was added to the other. Each vial was sonicated with a sonication probe at 50 amps for 4 minutes. Afterwards, 10 μL of drug solution was placed on a microscope slide, using wedge slide technique. Slides were imaged at 40 × magnification using EVOS microscope on a 4 mm × 4 mm area (total 108 images per sample). Another 10 μL of drug solution was placed on a flat silica substrate onto an SEM holder and sputtered with 7 nm of iridium. SEM images were taken using FEI Nova NanoSEM 230 ultra-high resolution, at the HMRI Microscope Core. Images were analyzed and particle size distribution determined using ImageJ and MATLAB®.
