Cell Migration Assay Protocol

Migration assays were performed using 8.0 μM polycarbonate transwell inserts (Corning) according to manufacturer's instructions. Briefly, the bottom wells were filled with 600 μL medium containing 10% bovine serum. A total of 1×105 cells in 100 μl serum-free medium were plated in the upper inserts and allowed to migrate for 24 hrs. Non-migrated cells were removed with a cotton swab, while cells that had migrated were fixed and stained with Hema-3 (Fisher Scientific) for counting. The fold change of cell migration was calculated by comparing the cells expressing miR-9 and HPV E6 with the cells expressing negative control RNA and empty vehicle vector, respectively.
