All chemicals used were of analytical grade and were purchased from the following providers: Sigma (St. Louis, MO, USA), Acros (Morris Plains, NJ, USA), Alltech (Deerfield, IL, USA), Fluka (Buchs, Switzerland), Supelco (Bellefonte, PA, USA), Prolabo, and VWR (Fontenaysous Bois, France). Endo-β-(1→4)-xylanase isolated from Aspergillus oryzae (Novazymes, EC 3.2.1.8, Shearzyme 2X, GH-10) that were used to hydrolyze xylans were obtained from Sigma.

Pectin Extraction from Apple Pomace

Pectins were isolated from dried apple pomace using sulfuric acid (pH 2.0, 85 °C, 3 h) or enzymatic preparations endo-β-1,4-xylanase (EC 3.2.1.8) from Trichoderma viride (Sigma/Aldrich Chemical Co., Germany, Cat. No. X3876) and endo-cellulase (endo-β-1,4-glucanase, EC 3.2.1.4) from Trichoderma viride (Sigma/Aldrich Chemical Co., Germany, Cat. No. C9422). Both enzymes were used at doses of 50 U/1 g of pomace, at pH 5.0, in 40 °C, for 10 h, according to a method described by Wikiera et al. [16 (link)]. The enzymatic preparations did not exhibit any additional off target activities [16 (link)].
Commercial apple pectin (Pcommercial) isolated with sulfuric acid obtained from Pektowin (Poland) was used as a reference. The primary chemical composition of pectin preparations used in the experiments is presented in the Table 5A,B.

Sugar Beet Arabinan Characterization

Sugar beet arabinan was purchased from Megazyme (Bray, Wirklow, Ireland). Genomic DNA Isolation Kit, PCR Amplification Kit, Gel Purification Kit, TA Cloning Kit and the restriction endonucleases were obtained from Fermentas (Fermentas-Life Science Technologies, Lithuania). All other chemicals including pNP-α-l-arabinofuranoside and endo β-1,4-xylanase were supplied by Sigma Chemical (St. Louis, MO, USA).

Enzymatic Hydrolysis of Lignocellulosic Biomass

The PHs as 1% (w/v) substrate were treated in 50 mM sodium citrate buffer (pH 4.8) supplemented with 0.01% (w/v) sodium azide. Each of the enzymes, celluclast (Novozymes, Denmark) and xylanase (endo-1,4-β-xylanase from Trichoderma longibrachiatum, Sigma-Aldrich, USA), were loaded with 30 FPU per gram of glucan and 300 international units (IU)/mL, respectively. All samples were completely suspended in rotary shaker at 200 rpm at 37°C for 48 h. All enzymatic hydrolysis experiments were performed in triplicate.

Enzymatic Hydrolysis of Lignocellulosic Biomass

Cellulose (Avicel PH101), xylan (beechwood) and endo-(1,4)-β-xylanase derived from Trichoderma longibrachiatum are purchased from Sigma-Aldrich, USA. Arabinoxylan (wheat) is purchased from Megazyme, and lignocellulose (Bambusa bambos) is collected from local area. Sodium acetate anhydrous, glacial acetic acid, 3,5-dinitrosalysilic acid, phenol, potassium sodium tartrate tetrahydrate, and sodium sulfite are purchased from Merck. D-xylose, sodium hydroxide (pellets), benzoic acid, thiourea, potassium bromide, and borax are purchased from SRL, Mumbai, India, and ortho-toluidine is purchased from Loba Chemie, Mumbai, India. All the chemicals used in this study are of analytical grade.
