Quantifying Th1 and CTL Cells in Lung Cancer

Check if the same lab product or an alternative is used in the 5 most similar protocols
To detect the proportion of Th1 and CTL cells, PBMCs from lung cancer patients and healthy donors were stimulated with 50 ng/mL phorbol myristate acetate (PMA; Sigma-Aldrich, CA, USA) and 1 µg/mL ionomycin (Sigma-Aldrich, CA, USA) for 2 h and then incubated for an additional 4 h in the presence of 1 µg/mL brefeldin-A (eBioscience, San Diego, CA, USA) at 37°C and 5% CO2. After that, cells were stained with human anti-CD3 and anti-CD8 mAbs (eBioscience), fixed, permeabilized, and stained with human anti-IFN-γ mAb (eBioscience) according to the Intracellular Staining Kit (Invitrogen, Carlsbad, CA, USA) instructions.

Quantifying T Cell Subsets in Lung Cancer

Check if the same lab product or an alternative is used in the 5 most similar protocols
To confirm the percentages of CD4+ and CD8+ T cells, 50 ng/ml phorbol myristate acetate (PMA; Sigma-Aldrich, California) and 1 μg/ml ionomycin (Sigma-Aldrich) were used to stimulate PBMCs from lung cancer patients and healthy donors for 2 h and then cells were incubated in the presence of 1 μg/ml brefeldin-A (eBioscience, San Diego) for another 4 h. Post stimulation, cells were then stained with human anti-CD3 and anti-CD8 mAbs (eBioscience), fixed, permeabilized and stained with a human anti-IFN-γ mAb (eBioscience) following the instructions of the Intracellular Staining Kit (Invitrogen, Carlsbad, CA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
