μCT Imaging of FLEx ACVR1(R206H) Mice

μCT imaging of FLEx ACVR1(R206H); CAG-cre/Esr1 mice was performed using a Quantum FX μCT Cabinet X-Ray System (PerkinElmer) with mice under isoflurane anesthesia and placed in a supine position for hind limb imaging. Scan parameters included 90 kV voltage, an 80 μm voxel size, and a 4.5-minute scan time. μCT imaging of Acvr1tnR206H/+; Tie2-Cre mice was performed using an IVIS SpectrumCT (PerkinElmer). All μCT images were taken using medium resolution acquisition mode (75 μm voxel size; estimated radiation dose of 132 mGy; 210 second scan time) with mice under isoflurane anesthesia. The μCT images were generated and HO volumes were quantified using 3D Slicer software (http://www.slicer.org), as previously described (6 (link), 12 (link)). Imaging of SCID hosts following cell transplantation was performed similarly.
