Polyvinylidene difluoride (PVDF) membrane is from Millipore. Transwells are purchased from Euroclone. Anti-Gal-1 antibody is from Cell Signaling, anti-β-actin and anti-Integrin-β1 antibodies are from Santa Cruz Biotechnology, anti-CD81 antibody is from BD Biosciences. The secondary antibodies enzyme horseradish peroxidase (HRP)–conjugated are from Santa Cruz Biotechnology. All materials for SDS-PAGE and Clarity™ Western ECL substrate are from Biorad. Culture-inserts (Dish 35 mm, high) are from ibidi®.

Antibody Procurement for HCV Research

Antibodies were purchased from the following sources: anti-CD81 antibody was from BD Pharmingen, mouse anti-actin antibody was from Sigma-Aldrich, and ISG15 and MxA antibodies were from Santa Cruz. HCV core, NS3, NS5A antibodies have been described elsewhere14 (link). HCV E2 antibody was a gift from Dr. Jean Dubuisson (Institut Pasteur de Lille). Tylophorine was purchased from Alexis Biochemicals. O859585, T298875, and Sofosbuvir were purchased from Toronto Research Chemicals and MedChemexpress.

Exosome Detection and Quantification

The exosome detection processing was as previously reported.9 (link) The 4 μm-diameter aldehyde/sulphate latex beads (Life Sciences, CA, USA) were incubated with purified anti-CD81 antibody (BD Biosciences, NJ, USA) for exosome binding. For allophycocyanin (APC) labeling, 50 μL anti-CD81 APC was added into each sample or matching isotype was added into control (BD Biosciences, NJ, USA). Samples were resuspended in 200 μL PBS buffer and analyzed by FACSCalibur system (BD Biosciences, NJ, USA). Ten thousand events were acquired using Cell Quest and the data were analyzed using FlowJo Software (Tree Star, Inc, Ashland, OR, USA).
