Bile Duct Ligation and BMSC Therapy

Check if the same lab product or an alternative is used in the 5 most similar protocols
Adult C57BL/6J male mice (8 weeks old) and aged mice (13–16 months old) were housed in a pathogen-free animal facility under a standard 12-hour light/dark cycle. Mice were given standard rodent chow and water ad libitum. Adult C57BL/6J male mice were divided randomly into four groups, and bile duct ligation in the mice was performed as previously described.39 (link) BMSCs obtained from C57BL/6J male mice (6 weeks old) were isolated and cultured according to an established protocol.40 (link) On post-operative day 23, 1 × 106 of Lenti.ctrl and Lenti.sg126 preconditioned BMSCs were injected via the caudal vein. Ad.sg126 mice and Ad.ctrl mice (C57BL/6J background) were generated using recombination Ad.sg126 or Ad.ctrl (Hanbio, Shanghai, China). Viral particles (1.0 × 1010) were reconstituted in 100 μL injected intravenously through the tail vein with BD Ultra-Fine Insulin Syringes. All procedures followed the Huazhong Agricultural University Guidelines for the Care and Use of Laboratory Animals.

Adenoviral Overexpression and Knockdown of Slc40a1

Check if the same lab product or an alternative is used in the 5 most similar protocols
Ad-Slc40a1 and Ad-Ctrl constructs were generated by Hanbio Biotechnology (China) using adenoviral vectors carrying Slc40a1. Ad-shSlc40a1 and Ad-shCtrl were provided by GeneChem Technology (Shanghai, China), in the form of adenoviral vectors carrying shRNA-Slc40a1.Protein expression was observed 48 hours after viral infection. The plasmid containing Steap4 and the negative control plasmid (OE-NC) were created and produced by Tsingke Biotech (China). Cardiomyocytes were transfected with the plasmid using Lipofectamine 3000 from Invitrogen (USA). Protein expression was measured 72 hours after transfection. The shRNA target sequences are shown in Table S3.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
