The largest database of trusted experimental protocols

The IC kit is a laboratory instrument designed for the analysis and separation of ionic compounds. It provides a solution for the detection and quantification of various ions, including anions and cations, in a wide range of samples. The core function of the IC kit is to perform ion chromatography, a technique used for the separation and measurement of ionic species.

Stimulation and Intracellular Cytokine Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Thymocytes were treated with 50 ng phorbol 12-myristate 13-acetate (PMA), 1 μg ionomycin and Brefeldin A for 4 h, before harvesting and staining intracellularly for IFN-γ, IL-4 and IL-17A using the eBiosciences IC kit (88-8824-00).

Flow Cytometry Analysis of Cytotoxic Cell Markers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were stained with the following antibodies from Biolegend: anti-human CD107a (H4A3), anti-human Granzyme B (GB11), and anti-human Perforin (#353312). Antibodies were coupled with APC and Pacific Blue fluorochromes. For intracellular cytokine production analysis, cells were either stimulated with PMA + Ionomycin + Brefeldin A for 4 h at 37 °C or incubated only with Brefeldin A and further stained with ebioscience IC kit according to the manufacturer’s instructions. Flow cytometry acquisition was performed on a LSR Fortessa (BD) and data was analyzed with FlowJo X software (Tree Star).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
