The analytical standard of PALBO was provided by Toronto Research Chemicals (Toronto, Canada, product N. P139900, lot N. 1-ZPK-125-1), RIBO hydrochloride (product N. M15373, lot N. 12195) and LETRO (product N. L6545, lot N. 0000028530) were supplied by Merck-Sigma Aldrich. Stable isotopically labeled internal standards D6-RIBO (product N. C4503, lot N. PO-ALS-18-007-B1), D8-PALBO (product N. C5108, lot N. SA-ALS-15-120-P1) and 13C2,15N2-LETRO (product N. C595, lot N. JA-ALS-18-104-P3) were synthesized by Alsachim (Illkirch Graffenstaden, France). Drug-free plasma/K-EDTA from healthy volunteers to prepare daily standard calibration curves and quality control samples (QCs) was provided by the transfusion unit of our institution.

Analytical Reagents and Standards

Cryoplateable Hepatocytes for NASH

Transporter Certified™ cryoplateable human hepatocytes were supplied by BioIVT along with all QualGro Media. Hepatocytes from NASH patients (BXW, GWD, IWM, and YNM) including male and female donors with nonalcoholic fatty liver disease activity score (NAS) scores ranging from 3 to 7 were used with 9-10 replicate samples analysed from each donor. For the sandwich-cultured human hepatocytes (SCHH), male and female donors were used (JHY, JPR, OSI, and XVN) with NAS scores of 0, with 6-8 replicate samples analysed from each donor for each condition (Table 1). HPLC grade chloroform, HPLC grade butanol (ButOH), LC-MS grade methanol (MeOH), and LC-MS grade isopropanol were purchased from Merck. LC-MS grade acetonitrile (ACN), Percoll, Dulbecco’s phosphate buffer saline (PBS), and formic acid were purchased from ThermoFisher Scientific. Ammonium carbonate was purchased from Rowe Scientific. Matrigel® was purchased from Bio-Strategy.

Quantitative Analysis of Isoprostanes

The standards for 8‐iso‐PGF2α and 8‐iso‐PGF2α‐d4 (internal standard) were purchased from Cayman Chemicals (Ann Arbor, MI), while the standard for PGF2α was obtained from Sigma-Aldrich (St Louis, MO). For solid‐phase microextraction (micro‐SPE), 96‐well Oasis® MAX μElution cartridges containing a water‐wettable reversed‐phase strong ammonium exchange mixed‐mode polymer, which is selective for acids and stable in organic eluents, were used. A Positive Pressure‐96 processor purchased from Waters was also used. Figure 1 shows the chromatograms of standards.

Cannabinoid Capsule Formulation Protocol
