Quantitative Assessment of Aortic Antioxidant Status

Check if the same lab product or an alternative is used in the 5 most similar protocols
Using the Rat Total antioxidant status (TAC) ELISA Kit (Catalog # MBS1600693, MyBioSource), the total antioxidant status was measured, and all the procedures were performed following the manufacturer’s instructions. The sandwich ELISA kit was utilized for the accurate quantitative detection of the total antioxidant status in the aortic tissue homogenate, and the reaction was terminated by the addition of the acidic stop solution and absorbance was measured at 450 nm.

Antioxidant Capacity and Phenolic Content Assays

Check if the same lab product or an alternative is used in the 5 most similar protocols
Determination of total phenolic contents, DPPH radical scavenging activity and FRAP ferric reducing antioxidant power assays were done as described9 (link). The total phenolic content was estimated using Folin-Ciocalteu method. In brief, 20 µL solution of the plant extract was mixed with 100 µL Folin-Ciocalteu reagent. At ambient temperature, the mixture was incubated for 5 min and then 80 µL of 7.5% Na2CO3 solution was added. The mixture was then left for 30 min in the dark and the absorbance was monitored using a microplate reader (Biochrom Asys UVM 340) at 735 nm. The results were presented as gallic acid equivalent (GAE) in mg/g of extract. Total antioxidant capacity (TAC) assay was determined using a commercially available TAC ELISA kit (MBS726896, my biosource, Inc., San Diego, CA, USA) according to the manufacturer’s instructions with ascorbic acid as the reference standard to estimate TAC as described10 (link).

Antioxidant Potential Evaluation

Check if the same lab product or an alternative is used in the 5 most similar protocols
The antioxidant potential of the extract was evaluated by determining its TAC using a TAC ELISA kit (MBS726896, my BioSource, Inc., San Diego, CA, USA) in accordance with the manufacturer’s instructions and compared to the standard antioxidant, ascorbic acid as previously described Sobeh et al. [3 (link)].

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
