Compound Preparation and Solubilization

The NIH Clinical Collections 1&2 (NIHCC 1&2, Evotec, San Francisco, CA) contain 727 compounds dissolved in 100% dimethyl sulfoxide (DMSO) at a concentration of 10 mM. Miconazole nitrate was obtained from Certa NV (Eigenbrakel, Belgium), meropenem from Astrazeneca (Zoetermeer, The Netherlands), econazole nitrate, ketoconazole, ciprofloxacin, and gentamicin sulfate from Sigma (Bornem, Belgium), oxiconazole nitrate from Santa Cruz Biotechnology (Heidelberg, Germany), and tobramycin from TCI chemicals (Tokyo, Japan). Azoles were dissolved in 100% DMSO (Sigma) and subsequently diluted in MilliQ water (MQ) (Millipore, Billerica, MA) for MIC determination, in physiological saline (PS, 0.9% w/v NaCl) for the checkerboard tests, for the tests with biofilms formed in 96-well microtiter plates (MTPs) and for the G. mellonella assay, or in GTSF-2 medium for the 3D organotypic human cell culture model [15 (link)]. Antibiotic stocks were made in MQ water or PS. All antibiotic solutions were filter sterilized (0.22 μm, Whatman, Dassel, Germany) and immediately used.

DABCYL is the abbreviated form of 4-(dimethylaminoazo)benzene-4-carboxylic acid and EDANS is the abbreviated form of 5-((2-Aminoethyl)amino)naphthalene-1-sulfonic acid.

Cyclodextrin-based Econazole Ocular Delivery

Characterization of Lymphoma Cell Lines

CB33, SUDHL4 and SUDHL6 cells provided by R. Dalla-Favera (Columbia University, NY) were maintained in IMDM (Life Technology), supplemented with 10% FBS (Gemini) and antibiotics. The HF1 follicular cell line provided by R. Levy (Stanford University, CA) was maintained in DMEM (Life Technology), supplemented with 10% FBS and antibiotics. Cells were tested negative for mycoplasma. Cells were not further authenticated. Antibodies: rabbit anti-MYC (XP) (Cell Signaling Technology); rabbit anti-FOXM1 and mouse anti-GAPDH (SantaCruz); rabbit anti-HMGA1, anti-ATF5, anti-NFYB, mouse anti-TFDP1 (Abcam). Alprostadil, Clemastine, Cytarabine and Troglitazone (Tocris), Econazole nitrate and Promazine hydrochloride (Sigma) were reconstituted in DMSO (Sigma).

Screening Library for Parasitic Worms

Based on a comparative genomics analysis, we previously proposed a screening set for parasitic worms, of 817 drugs approved for human use [11 (link)]. Taking into account price, availability and chemical class, 409 compounds were obtained from Sigma-Aldrich in 10 mM dimethylsulfoxide (DMSO) solutions, and stored at -20°C (S1 Table and Fig 1). For in vivo testing, we obtained high-purity compounds from Sigma-Aldrich: econazole nitrate (catalogue number E0050000), prazosin hydrochloride (BP399), flunarizine dihydrochloride (F0189900), butoconazole nitrate (1082300), felodipine (BP777), cyproheptadine hydrochloride (1161000), terfenadine (T00710000), pimozide (BP682 or P1793), nicardipine hydrochloride (1463224 or N7510), mebendazole (1375502), as well as Tween 80 (P6474) and ethanol (PHR1373) for dissolving the drugs.
