Mitochondrial Potential and ROS Assays

Check if the same lab product or an alternative is used in the 5 most similar protocols
An enhanced mitochondrial membrane potential assay kit with JC-1 (Beyotime) was used to examine BMSCs treated with H2O2 and cisplatin. A Reactive Oxygen Species (ROS) Assay Kit (Beyotime) was used to detect the antioxidative capacity of cells. Both JC-1 and ROS assays were observed under a fluorescence microscope.

Quantifying Mitochondrial Membrane Potential

Check if the same lab product or an alternative is used in the 5 most similar protocols
ΔΨm was determined by an assay kit with JC-1, according to the manufacturer's instructions (Beyotime Institute of Biotechnology, Haimen, Jiangsu. China). JC-1 stains the mitochondria in cells with a high ΔΨm by forming red fluorescence J-aggregates, whereas in cells with depolarized mitochondria, JC-1 is present as green fluorescent monomers. In this way, mitochondrial depolarization was determined by a decreased ratio of red-to green fluorescence intensity by a fluorescence microscope (Zeiss Inc., Jena, Germany).

Assessing Mitochondrial Membrane Potential in GCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Briefly, to examine the MMP of GCs in all treated groups, the mitochondrial membrane potential assay kit with JC-1 (Beyotime, China) was used according to the manufacturer’s instructions. Enzymatic digestion using trypsin was used to harvest the GCs and washed with warm PBS. After collection, the MMP assay kit with JC-1 (Beyotime, China) was used to stain GCs. Flow cytometry was used to count labeled cells using a fluorescence activating cell sorter (FACS) and a Calibur flow cytometer (BD Biosciences, CA, USA). The data was analyzed using FlowJo software (version Win 64-10.4.0).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
