ACY1215 was purchased from Chemietek (Indianapolis, IN). siRNA targeting human HDAC6 (SI02663808 [siHDAC6_A], SI02757769 [siHDAC6_B], SI03058706 [siHDAC6_C], and SI04438490 [siHDAC6_D]), Notch1 (SI00119035), and AllStars Negative Control siRNA (SI03650318) was purchased from Qiagen (Valencia, CA, USA). Transfections were conducted using the Lipofectamine 2000 Transfection Reagent following the manufacturer's protocol (Invitrogen).

Enhancing ACY-1215 Bioavailability

The reagent 2-(diphenylamino)-N-(7-(hydroxyamino)-7-oxoheptyl) pyrimidine-5-carboxamide (ACY-1215) was purchased from Chemietek (Indianapolis, IN). ACY-1215 (ACY) was first dissolved in DMSO (Sigma–Aldrich, St. Louis, MO) and then in culture medium (1–2 µM) for cell culture. Because PEG-400 enhances the bioavailability of ACY, it was dissolved in PEG-400 (Sigma–Aldrich) and diluted with sterile water. ACY was administrated to mice by gavage.

The following primary antibodies were purchased from the following companies: Santa Cruz (Dallas, TX; HDAC6 [H-300], HIF-1α), Abcam (Cambridge, MA; type-1 collagen), Sigma-Aldrich (α-SMA, acetylated α-tubulin), Cell Signaling Technology (Danvers, MA; β-actin, Smad2, phosphorylated Smad2, Smad3, phosphorylated Smad3, Akt, phosphorylated Akt (Ser473), PHLPP, Erk, phosphorylated Erk, p38, phosphorylated p38, HDAC6 [for the mouse samples], α-tubulin, LC3B). Secondary antibodies used were anti-mouse or anti-rabbit IgG HRP-linked antibodies (Cell Signaling Technology).

Histone Deacetylase Inhibitor Effects on Xenograft Tumor Growth

Cells were treated with 20μM Tubacin (Sigma), 10-20μM Tubastatin A (ChemScene), and 2μM ACY1215 (ChemieTek) or 1μg/ml Trichostatin A (Sigma) for 2-12h, and disrupted in a PTY buffer (35 (link)) and processed for WB or IP. b) Xenograft studies. Compound administration began 1 week after the injection of the tumor cells into the mammary fat pad of the mice. Tubastatin A was dissolved to 70mg/ml in DMSO and brought to 10mg/ml with PBS. A 40 mg/kg/dose was administered via intra peritoneal injection once a day for 5 days/week for a total of 2 weeks (36 (link)). For the lung metastasis studies, Tubastatin A was administered in the same manner as above, starting 24 h after the tumor cell tail vein injections. ACY-1215 was dissolved to 40mg/ml in DMSO and brought to 24mg/ml with saline. A 75 mg/kg/dose was administered via intra peritoneal injection once a day for 5 days/week for a total of 2 weeks (37 (link)).
