Immunohistochemical Analysis of Xenograft Tumors

Xenograft tumors were fixed with 10% formaldehyde and embedded in paraffin, followed by being sliced, dewaxed, and hydrated. Antigen retrieval was performed using 0.01 M citrate buffer. H2O2 solution (3%) was applied to incubate sections for 15 minutes at room temperature. Goat serum (5%) was added onto sections for 30 minutes incubation at room temperature. Thereafter, sections were sequentially subjected to incubation with primary antibody (1:100, mouse anti-Ki67 and anti-PTEN, Geneseed Biotech, Guangzhou, China) overnight at 4°C and with HRP labeled secondary antibody (1:300, Boster, Wuhan, China) for 30 minutes at room temperature. After being stained with diaminobenzidine (DAB) and hematoxylin, sections were sealed with neutral resin. The positive signals of Ki67 and PTEN were observed under a microscope (Olympus, Tokyo, Japan).

Evaluating Cell Proliferation via Ki-67 Immunostaining

The tumor tissues were fixed with 4% paraformaldehyde, embedded in paraffin, and cut into 5-μm-thick sections. Then the paraffin sections were incubated with primary antibodies against Ki-67(1:100, Geneseed Biotech, Guangzhou, China) overnight at 4°C, followed with HRP labeled secondary antibody (1:300, Boster, Wuhan, China) for 30 minutes at room temperature.
