Plasma and Urine Creatinine and Urea Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The concentrations of creatinine in the plasma and urine were assayed using the kit (K625-100, BioVision, California, USA). The concentration of urea in the plasma was determined using the kit (K375-100, BioVision, California, USA).

Quantifying Albumin and Urea Secretion in Spheroids and Organoids

Check if the same lab product or an alternative is used in the 5 most similar protocols
To determine the levels of albumin secreted over time by spheroids and organoids, co-culture media were collected from perfusion channels and graft chambers (day 3, 5, and 7 for spheroids, day 7, 14, and 21 for organoids) and analyzed using the Human Albumin ELISA Quantification kit (Bethyl Laboratories Inc., E80-129, sample dilution 1:125).
Urea production was determined by a colorimetric assay kit (Biovision, #K375-100, sample dilution 1:5) following the manufacturer’s protocol.

Metabolite Profiling of Hepatocyte Supernatants

Check if the same lab product or an alternative is used in the 5 most similar protocols
Levels of glucose, glutamic acid and urate from supernatants were measured using AmplexRed-based assays (Invitrogen: A22189, A12221 and A22181) according to manufacturer's directions for the Qubit fluorometer. Urea levels were measured using a colorimetric assay kit (Biovision, K375-100) according to manufacturer's directions. Following the assays, the background level of each measured metabolite in fresh WEM or WEM with theophylline was averaged (n=2) and subtracted from the individual respective supernatant samples. Endpoint concentrations were determined by adding back the average baseline concentration of fresh media without theophylline. Endpoint concentration changes between theophylline (n=4) and control treated samples (n=4) were determined for each metabolite using an unpaired two-sided Student's t-test. Statistical significance was determined independently for rat and human hepatocyte experiments by correcting for multiple hypothesis testing across the four metabolites and applying a FDR-adjusted q-value threshold of 0.05.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
