Quantifying Alveolar Cytokine Release

To quantify cytokines released in the alveolar in vitro model, ELISA assays were performed. We quantified IL8 with SimpleStep ELISA® Kit (Abcam), TGF-β (Abcam) and IL1β with human IL-1β/IL-1F2 Immunoassay (R&D Systems) was titered using a commercial enzyme-linked immunosorbent assay kit (human IL-1β/IL-1F2 Immunoassay, R&D Systems) following the manufacturer’s instructions and the results were expressed as pg ml-1. All determinations were measure in same session. For IL8 quantification in BAL of COVID-19 patients we referred to already published quantifications (10 (link)).

Serum Cytokine Quantification Using ELISA

Serum L-1β and TNF-α concentrations were measured using the Quantikine HS ELISA, Human IL-1β/IL-1F2 Immunoassay and Human TNF-α Immunoassay (R&D Systems) according to the attached protocol. Referring to the attached protocol, the sensitivity range for IL-1β for 25 assays was 0.014–0.063 pg/mL and the TNF-α sensitivity range for 23 assays was 0.011–0.049 pg/mL. The optical density (450 nm) after the color reaction was measured using a TriStar LB941-vTi Microplate Reader (Berthold Technologies).
