Trace Metal Analysis of Samples

Composition analysis was carried out at the Analytical Chemistry Services of the Iowa State University Veterinary Diagnostic Laboratory. Samples were analysed for copper and zinc content using an Inductively Coupled Plasma Mass Spectrometry (Plasma Quant MS Elite, Analytik Jena Inc. Woburn, MA, USA) in CRI mode with hydrogen as the skimmer gas. Standards were obtained from Inorganic Ventures (Christiansburg, VA). 15 mL centrifuge tubes and trace mineral grade nitric acid were purchased from Fisher Scientific (Pittsburgh, PA). Samples were partitioned into 15 mL tubes and digested in 70% nitric acid at 70 °C in a dry heat bath for 2 h. After digestion, samples were diluted using 1% nitric acid. Samples with elevated copper content occasionally needed an additional dilution made using 1% nitric acid and were analysed by ICP-MS after the dilution. For copper and zinc analysis, yttrium was used as internal standards for the ICP-MS analysis.

Elemental Analysis of Mouse Colon Samples

Colon samples were carefully excised from mice designated for biochemical and neurochemical analyses and weighed prior to sample preparation. Samples were analyzed for cadmium (Cd), Ca, cromium (Cr), cobalt (Co), copper (Cu), iron (Fe), Mg, Mn, molybdenum (Mo), phosphorus (P), potassium (K), selenium (Se), sodium (Na), and zinc (Zn), using ICP-MS (Analytik Jena, Inc.) in CRI mode with hydrogen as the skimmer gas. Standards for elemental analyses were obtained from Inorganic Ventures, and digestion vessels, trace mineral grade nitric acid, and hydrochloric acid were obtained from Fisher Scientific. Briefly, samples were digested in 70% v/v nitric acid at 60°C for 12h . Preweighed samples were transferred to 15-mL tubes, and 0.25mL of 70% nitric acid was added. All samples were digested overnight at 60°C. After digestion, all samples were diluted to 5mL using 1% v/v nitric acid with 0.5% v/v hydrochloric acid and then analyzed by ICP-MS. For quality control, bismuth (Bi), scandium (Sc), indium (In), lithium (Li), yttrium (Y) and terbium (Tb) were used as internal standards for the ICP-MS.

samples were transferred to 15 mL tubes and 0.25 mL of 70% nitric acid was
added. All samples were digested overnight at 60˚C. After digestion, all
samples were diluted to 5 mL using 1% nitric acid with 0.5% hydrochloric acid
and then analyzed by ICP-MS. For quality control, Bi, Sc, In, Li, Y, and Tb were
used as internal standards for the ICP-MS.
