Quantification of Bile Acids and Metabolites

Cholic acid 7-sulfate (CA-S) and 3′-sialyllactose were purchased from Cayman (Biomol GmbH, Hamburg, Germany). Sulfate (S) conjugates of CDCA, UDCA, and LCA were synthesized according to Donazzolo et al. (Donazzolo et al., 2017 (link)) and structures were verified by MS/MS and NMR spectroscopy.
Milli-Q water (18.2 MΩ) was derived from a Milli-Q Integral Water Purification System (Billerica, MA, United States). Acetonitrile (ACN; LiChrosolv®, hypergrade for LC-MS), methanol (LiChrosolv®, hypergrade for LC-MS) and ammonium acetate (NH4Ac) were obtained from Merck (Darmstadt, Germany). Glacial acetic acid was purchased from Biosolve (Valkenswaard, Netherlands) and formic acid from Honeywell Fluka™ (Morristown, NJ, United States).

Wy was purchased from Chemsyn Science Laboratories (Lenexa, KS) and mixed into a custom grain-based diet by Research Diets (New Brunswick, NJ). CA, deoxycholic acid (DCA), CDCA, taurocholic acid (TCA), taurodeoxycholic acid (TDCA), taurochenodeoxycholic acid (TCDCA), tauroursodeoxycholic acid (TUDCA), and taurohyodeoxycholic acid (THDCA) were purchased from Sigma-Aldrich (St. Louis, MO). βMCA, ω-MCA, tauro-α-muricholic acid (TαMCA), tauro-β-muricholic acid (TβMCA), and d5-TCA were obtained from Toronto Research Chemicals (North York, ON, Canada). All other reagents were of the highest grade commercially available.
