Whole Blood RNA Sequencing Protocol

Subjects were enrolled during their ED stay, at which time peripheral whole blood was collected in QIAGEN PAXgene Blood RNA tubes, then processed according to manufacturer instructions, and stored at −80ºC. Total RNA was extracted with QIAGEN miRNA kit following all manufacturer instructions. Total RNA quantity and quality were assessed using the Nanodrop spectrophotometer (ThermoScientific) and Agilent Bioanalyzer with RNA 6000 Nano kit. Libraries were prepared using NUGEN Universal Plus mRNA Seq Kit with AnyDeplete globin and rRNA depletion (Tecan Life Sciences) and sequenced using Illumina NovaSeq 6000 S4 flow cell at 150 bp paired-end (PE) with an average of 40M read pairs per RNA sample.
