Cholangiocarcinoma Tissue and Cell Line Protocol

The human tumor and paired para-tumor tissues and ICC specimens of the validation cohort were obtained from ICC patients undergoing hepatectomy at Renji Hospital affiliated to Shanghai Jiaotong University School of Medicine. Patient samples were obtained following informed consent and protocols that were approved by the ethical review committee of the World Health Organization's Collaborating Center for Research in Human Production (authorized by Shanghai Municipal Government). The cholangiocarcinoma cell line RBE, HuCCT1 and FRH-0201 were purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China).

Culturing Human Cell Lines

Culturing Hepatic and Biliary Cell Lines

The human intrahepatic biliary epithelial cell line HIBEpiC, IHCC cell line RBE, PHCC cell line QBC939 and FRH0201 were purchased from Cell Bank of the Chinese Academy of Sciences (Shanghai, China). IHCC cell line HuCCT1 was purchased from RIKEN Bioresourse Center (Koyadai, Japan). HEK 293 Phoenix ampho packaging cells were purchased from the American Type Culture Collection. All cell lines were cultured in DMEM medium or RPMI 1640 medium supplemented with 10% fetal bovine serum and penicillin/streptomycin. All cells were used no more than 6 months after purchase for the experiments described herein.

Cell Line Authentication and Inhibitor Screening

Human PHCCA cell lines QBC-939 and FRH-0201 and IHCCA cell lines RBE and HCCC-9810 were all purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). All cell lines were maintained in DMEM (Gibco, Grand Island, New York, USA) supplemented with 10% fetal bovine serum (Gibco) and penicillin/streptomycin (HyClone) in 5% CO2. All cell lines were authenticated using short tandem repeat (STR) analysis, and the databases of the Chinese Academy of Sciences and American Type Culture Collection were used as references.
The inhibitors AACOCF3 (APExBIO) and celecoxib (Selleck, Houston, TX, USA) were used. All other agents without special instructions were purchased from Sigma.
