Standardized Zearalenone Metabolite Measurements

The solid standards of ZEN, ZAN, β-ZAL, α-ZAL, β-ZEL, α-ZEL and C13-ZEN were obtained from Sigma (St. Louis, MO, USA) and stored at −20 °C before use. The standards of ZEN-14GlcA, β-ZEL-14GlcA, α-ZEL-14GlcA were provided by the lab of Dr. Bart Huyberchts at CODA-CERVA. All organic solvents, salts and acid were analytical or HPLC grade. Acetonitrile, methanol, hexane, and isopropanol were purchased from Honeywell. Ammonium acetate and formic acid were purchased from Sigma. Milli-Q-quality water (Millipore, Billerica, MA, USA) was used throughout the experiments. C18-columns were obtained from Agela Technologies (Tianjin, China).

Fabrication of SU-8 Microfluidic Devices

Single-side polished 100 mm diameter <100> orientation silicon wafers were purchased from Siegert wafer GmbH (Aachen, Germany). SU-8 negative photoresist (SU-8 50) and developer (mr-DEV-600) were purchased from Microresist Technologies GmbH (Berlin, Germany). AZ 5412E, AZ 15nXT (450 CPS), MIF 312 developer and TechniStrip® NI555 was purchased from Microchemicals GmbH (Ulm, Germany). OmniCoat and PMGI are purchased from MicroChem Corp. (Westborough, MA, USA). Buffered HF and isopropanol were purchased from Honeywell International Inc., (Kuopio, Finland). Methanol, formic acid, and verapamil for mass spectrometry were purchased from Sigma Aldrich (Espoo, Finland). Polymer sheets used as carriers for bonding purposes were purchased from Arron (Helsinki, Finland). Chrome glass masks for ESI chips were acquired from ML&S GmbH (Greifswald, Germany) and film masks for metalized microchannels were from Micro Lithography Services Limited (Chelmsford, UK). All the reagents for clean room purposes are VLSI grade and for mass spectroscopy they are LC-mass spectrometry (MS) grade.

Acoustic Vibration Substrate Analysis

A 100-Hz acoustic vibration was generated from a programmed Arduino board (serving as a sinusoidal waveform generator) connected to a 45-mm 0.2-W super-thin Mylar speaker, as previously employed [13 (link)]. Beef extract, yeast extract, and peptone were from HiMedia (Maharashtra, India). NaCl was from Ajax Finechem-Thermo Scientific (Rockford, IL, USA). CHCl3 was from RCI Labscan (Bangkok, Thailand). MeOH and ammonium hydroxide (25%) were from Merck (Rahway, NJ, USA). Isopropanol was obtained from Honeywell (Charlotte, NC, USA). Formic acid was from Sigma-Aldrich (Milwaukee, WI, USA). Rhamnolipids (R90; 90%) were obtained from AGAE Technologies-Sigma-Aldrich (Milwaukee, WI, USA), and sodium oleate from Tokyo Chemical Industry (Tokyo, Japan). All chemicals were reagent grade or liquid chromatography grade or better as appropriate and used as supplied.

RNA Isolation and cDNA Synthesis Protocol

Total RNA isolation was performed via phenol-chloroform extraction. TriFast was added to the cells and incubated for 5 min at room temperature. A total of 100 μL chloroform per 500 µL Trifast was added (Carl Roth, Karlsruhe, Germany), incubated for 10 min in RT, and centrifuged at 14,000× g for 10 min at 4 °C. The upper aqueous phase, which contains the RNA, was collected carefully into a new tube containing 250 μL isopropanol (Honeywell, Charlotte, NC, USA). After incubation overnight at −20 °C, centrifugation with the same conditions as before was performed. The supernatant was discarded, and the RNA pellet was washed twice with 70% ethanol, followed by centrifugation. After discarding the supernatant, the tubes were left open to evaporate the remaining ethanol residues. The pellet was resuspended with DEPC H2O. The cDNA was synthesized using the First Strand cDNA Synthesis Kit (Thermo Fisher Scientific, Sindelfingen, Germany), according to the manufacturer’s instructions.

Purification and Characterization of HIV-1 Antigens

The Galanthus nivalis lectin (GNL)- and Q-sepharose-purified HIV-1 CO6980v0c22 gp145 reference material (RM) expressed in CHO-K1 cells was obtained from Advanced Bioscience Laboratories (ABL Inc.). HIV-1 SF162 gp140 recombinant protein produced in HEK 293T cells [23 (link)–26 (link)] was provided by the NIH AIDS Reagent Program, Division of AIDS, NIAID, NIH. Acetonitrile, isopropanol, methanol, n-propanol, trifluoroacetic acid (TFA), and LC-MS grade water, were purchased from Honeywell (Charlotte, NC, USA). The Dulbecco’s Phosphate-Buffered Saline (D-PBS, Corning, Corning, NY, USA) 1X without calcium and magnesium was obtained from VWR. CHO-K1 PC-2 spent medium was obtained from CDI Laboratories.

Quantification of ALMS1 mRNA Levels

After transfection, cells were incubated for 24h followed by 48h of starvation. Total RNA from control and ALMS1 deficient cells were isolated using Trifast (30–2010, VWR), Chloroform (1.02445, Merck), Isopropanol (34,965, Honeywell) and 75% Ethanol (Ethanol absolute, 1.00983, Honeywell). CDNA was synthesized using reverse transcriptase (M1705, Promega), Hexamers (C1181, Promega), Oligo Nucleotide (C118A, Promega). SYBR Green master mix (1725274, BioRad) and BioRad qPCR Cycler was used to perform qPCR amplification and measurement. Primers and qPCR conditions are listed in the supplement table.

Analytical Characterization of Fuel Additives

FFO components were supplied by BP Castrol (Pangbourne, UK) and prepared in‐house to resemble an FFO according to the formulation reported below. n‐Decane, 1‐decene, n‐dodecane and 1‐dodecene were all purchased from Fisher Chemicals (Loughborough, UK) and diluted 1 in 10 using toluene for analysis. n‐Decane and n‐dodecane were of 99% purity, whilst 1‐decene and 1‐dodecene were of 96% purity (remainder isomers).
