Quantifying Protein Expression by Western Blot

Check if the same lab product or an alternative is used in the 5 most similar protocols
Protein levels of GFP, dnRhoA, and β-Actin were assessed using Western Blot, following previously described protocols.27 (link) Briefly, 5 μg of protein were separated on pre-cast SDS-PAGE gels (Bio-Rad) and transferred onto Immobilon IP membranes (Millipore) before probing for GFP (1:2500, Abcam, #ab6556), RhoA (1:500, #MA1-134, Thermo Fisher Scientific), β-Actin (1:2500, #ab170325, Abcam), and GAPDH (1:50,000, #PA1-9046, Thermo Fisher Scientific). Primary antibodies were detected using IRDye 800CW goat anti-mouse (#925-332210), IRDye 680RD donkey anti-rabbit (#925-68073), or IRDye 800CW donkey anti-goat (#926-32214) secondary antibodies (1:15,000, LI-COR). Blots were visualized on an Odyssey Infrared Imaging System (LI-COR). Blots were stripped with NewBlot PVDF Stripping Buffer (LI-COR) and reprobed once to allow for detection of all four proteins on the same blot.

Quantitative Immunoblotting of GFP Variants

Check if the same lab product or an alternative is used in the 5 most similar protocols
Bacterial cells expressing GFP variants, either unfused or fused to proinsulin, were harvested and processed for SDS-PAGE as previously described.10 (link) Immunoblotting was then performed8 (link) with a 1:2500 dilution of a rabbit anti-GFP polyclonal antibody (Abcam, catalog number ab6556) followed by a 1:3000 dilution of an Alexa Fluor 488-conjugated goat anti-rabbit secondary antibody (Thermo Fisher, catalog number A-11008). Signals were detected using a LI-COR Odyssey CLx system.

Immunohistochemical Analysis of DLBCL

Check if the same lab product or an alternative is used in the 5 most similar protocols
Sections of paraffin-embedded formalin-fixed spleens (4 µm) were deparaffinized and stained with H&E or with specific antibodies to BCL2 (Abcam ab182858, 1:8000), GFP (Abcam ab6556, 1:1000), Ki-67 (Thermo Scientific RM9106, 1:100), or MYC (Abcam ab51154, 1:50). Of note, BCL2 antibody was able to detect both human and murine BCL2 as reported. Secondary antibodies against primary Fc were conjugated with horseradish peroxidase enzyme and developed by DAB color substrates, followed by counterstaining with hematoxylin. All slides were viewed under a light microscope and scanned using either a Leica Aperio CS2 whole-slide scanner at ×20 magnification (for an expert pathologist evaluation and confirmation of the high grade DLBCL phenotype), or a Nikon NIS Elements Microscope Imaging at ×20 and ×100 magnification (for visualization of tumors at a greater resolution and magnification).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
