Evaluating Compound Metabolic Stability

Check if the same lab product or an alternative is used in the 5 most similar protocols
The metabolic stability of selected hits was tested using human, bovine, horse and mouse liver microsomes (Corning) of phase-I (CYP450 and NADPH dependent enzymes) and phase-II (UGT enzymes) metabolism as described before (Hulpia et al., 2019a (link)). In short, samples were collected after 0, 15, 30 and 60 min and the corresponding loss of parent compound was determined using ultra-performance liquid chromatography (UPLC) (Waters AquityTM) coupled with tandem quadrupole mass spectrometry (MS2) (Waters XevoTM), equipped with an electrospray ionization (ESI) interface and operated in multiple reaction monitoring (MRM) mode.

Targeted UPLC-MS/MS for Compound Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Chromatography was performed on a Waters Acquity UPLC system coupled to a Waters Xevo TM dependent acquisition where the 8 most intense ions detected in the MS spectrum were selected as MS/MS precursors. Typically, the threshold for MS/MS targeting was set to 50 with targeted ions subsequently excluded from targeting for 15 seconds. MS/MS scans were obtained for selected ions with CID fragmentation, isolation width of 2.0, normalized collision energy of 36 V, activation Q of 0.250, and activation time of 30 ms.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
