Cisplatin (DDP) was purchased from Aladdin (Shanghai, China). NADPH and 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB) were obtained from Biosharp (Guangzhou, China). Aurothioglucose (ATG) was purchased from WAKO (Tokyo, Japan). Glutathione reductase (GR) and iodoacetamide (IAM) were purchased from Sigma-Aldrich (St. Louis, MO, USA). RIPA lysis buffer was obtained from Beyotime Biotechnology (Shanghai, China). Selenium-deficient (SD, 0.01 mg/kg), selenium-normal (SN, 0.21 mg/kg), and selenium-enriched (SE, 1.01 mg/kg) diets were manufactured by Medicience (Nanning, China). All other reagents were obtained from the Sigma-Aldrich Chemical Co. (St. Louis, MO, USA) or Fisher Scientific Ltd. (Waltham, MA, USA). The pure recombinant wild-type rat TrxR1 protein, and anti-Trx1 and anti-TrxR1 antibodies were obtained from IMCO Corporation, Stockholm, Sweden (www.imcocorp.se accessed on 1 January 2020). Other antibodies were obtained from Proteintech (Wuhan, China). C57BL/6j mice were provided by Ensiweier (Chongqing, China).

Western Blot Profiling of NPAS2

Cells were collected and lysed with RIPA buffer to extract protein samples. The proteins were run on SDS-PAGE gels and transferred to PVDF membranes. The membranes were blocked with 5% non-fat skim milk, followed by incubation with primary antibodies at 4 °C overnight and incubated by secondary antibody for 1 h at room temperature. Ultimately, the specific bands were detected using ECL chromogenic solution. The intensity of bands was performed using Image J software. Anti-NPAS2 antibody was purchased from NOVUS (USA), other antibodies were commercially purchased from Proteintech (Wuhan, China).

Oxidative Stress in Trophoblast Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
HTR-8/SVneo cells (provided by Dr. Charles Graham, Queen's University of Canada) at the logarithmic growth stage were cultured in a 37°C, 5% CO 2 incubator plated on a six-well culture plate at a density of 1 × 10 5 cells per well. After 24 hours, H 2 O 2 (Sigma-Aldrich, USA) was added at concentrations of 300 µmol/L and 3hours to the culture to establish an oxidative stress model of HTR-8/SVneo cells (Model). Cells in the experimental group were incubated with gradient diluted BHD water extracts (1 mg/mL, 5 mg/mL, 10 mg/mL, 20 mg/mL) for 48 h, and cells without any treatment were used as the control group (Control). The morphology of the cells was observed under an inverted microscope.
PTGFR was obtained from Abcam (Cambridge, UK), and other antibodies were obtained from Proteintech (Chicago, USA). PTGFR (ab126709, 1:2000), AGTR1(25343-1-AP, 1:500), OXTR (23045-1-AP, 1:1000), GAPDH (10494-1-AP, 1:5000).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
