Multiparameter Flow Cytometry Immunophenotyping

Surface marker staining was performed in DPBS with 1% FBS using antibody concentration recommended by the supplier. Intracellular staining was performed following the supplier's protocol using Fix & Perm (BD Biosciences). Flow cytometric analysis was performed on the LSR II and cell sorting on the ARIA II (both BD Biosciences), both equipped with fur lasers. All populations studied were devoid of dead cells based on propidium iodide negativity and of doublets based on FSC-A FSC-W ratios. Comprehensive Immune Cell Profiling

Isolated PBMCs were stained with antibodies against human CD132 (IL-2RG), CD3, CD4, CD8, CD14, CD19, CD56, TCRαβ, and TCRγδ (Miltenyi Biotec, Bergisch Gladbach, Germany). Samples were incubated with the antibodies for 10min at room temperature in the dark and were read by flow cytometer (BD FACS Calibur, BD Biosciences, Franklin Lakes, USA). Data were carried out using FlowJo 10.7.2 software (BD Biosciences, Franklin Lakes, NJ, USA).
