Synthesis of PD-L1/CTLA4 Conjugates

The synthesis was performed as described previously for PlGF-2123–144 conjugates (16 (link)). Rat anti-mouse PD-L1 (clone: 10F.9G2, Bio X Cell) and hamster anti-mouse CTLA4 (clone: 9H10, Bio X Cell) were incubated with 15 eq. of sulfo-SMCC for 40 min at room temperature (RT). Excess sulfo-SMCC was removed using a Zeba spin desalting column (Thermo Fisher Scientific). >10 eq. of CBD protein (the VWF A3 domain, with an C-terminal cysteine residue) was then added and reacted for 1 hour at RT.

Modulating Tumor Immunity via CTLA4 and PD-L1

8- to 12-week-old female mice were injected subcutaneously with 100μl PBS containing 5×105 B16ova melanoma cells into the left, ventral flank. Intraperitoneal injection of 200μl PBS containing either 0.4mg of a Syrian hamster anti-mouse CTLA4 (Clone UC10-4F10-11; BE0131), 0.5mg rat anti-mouse PD-L1 (Clone10F.9G2; BE0101), 0.4mg Syrian hamster IgG (BE0087) or 0.5mg rat IgG2b (Clone LTF-2; BE0090) (all from BioXCell, USA) was performed every 3 days starting from day 0 of B16ova challenge until day 18.
Tumor volume was calculated according to the following equation: V = π/6*length*width2. For survival analysis, mice with tumors exceeding the length limit of 15 mm were sacrificed and counted as dead.

Rat anti-mouse IFN-γ (clone: XMG1.2), rat IgG1 isotype control (clone: HRPN), rat anti-mouse CD25 (clone: PC-61.5.3), rat anti-mouse PD-L1 (clone: 10F.9G2), rat IgG2b isotype control (clone: LTF-2) and mouse IgG1 isotype control (clone: MOPC-21) antibodies were from Bioxcell. 3-Methylcholanthrene (MCA) was from Sigma-Aldrich. MCA-205 cell line was kindly provided by Dr. Tomasz Zal (MD Anderson Cancer Center, TX).
