Short hairpin tau and rna ctrl vectors

Manufactured by Santa Cruz Biotechnology

The Short hairpin Tau and RNA Ctrl vectors are genetic constructs designed for targeted gene silencing. These vectors contain short hairpin RNA (shRNA) sequences that can be used to suppress the expression of the tau protein. The RNA Ctrl vector serves as a control for the shRNA-mediated knockdown experiments. The core function of these vectors is to provide a tool for researchers to investigate the role of the tau protein in cellular processes.

pcDNA3-Tau4R and Tau-GFP have been described elsewhere [14 (link),23 (link),24 (link)]. pimEJ5GFP was a gift from Jeremy Stark (Addgene plasmid # 44026) (Watertown, MA, USA) [25 (link)]. pDRGFP (Addgene plasmid # 26475) and pCBASceI (Addgene plasmid # 26477) were a gift from Maria Jasin [26 (link)]. Short hairpin Tau and RNA Ctrl vectors were purchased from Santacruz. Doxorubicin, cisplatin, oxaliplatin, 6-thioguanine, and hypoxanthine were purchased from Sigma-Aldrich (Saint-Louis, MO, USA), and bleomycin from Calbiochem (from Sigma, St. Louis, MO, USA). Cells were exposed to ionizing radiation (IR) using an X-ray machine (Clinac23X, Varian Medical Systems, Palo Alto, CA, USA).

pGEX vectors encoding Tau and Tau-deletion mutants and pGEX-CBP (aa 1202–1848) fused to GST tag were a kind gift from J.C. Lambert (INSERM U1167, Lille, France) and C. Smet-Nocca (UMR8576, Villeneuve d’Ascq, France), respectively. GFP-HP1β was a gift from Tom Misteli (Addgene plasmid # 17651). pGL4.31(luc2P/GAL4 UAS/Hygro) was purchased from Promega. Short hairpin Tau and RNA Ctrl vectors were purchased from Santacruz. pcDNA3-Tau4R has been described elsewhere (Lippens et al., 2004 (link); Chauderlier et al., 2018 (link)). GAL4-Tau4R was obtained by inserting an in-frame Tau1N4R (referred to as Tau4R) cDNA isoform into the pM GAL4 DNA-BD cloning vector (Clontech). Purified histones and recombinant histones were obtained from EpiCypher and New England Biolabs, respectively. TSA and BIX 01294 (Sigma) were reconstituted in dimethylsulfoxide and Tetracycline (Sigma) in ethanol.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
