Transverse Aortic Constriction in Mice

Transverse aortic constriction (TAC) was constructed as previously described. Briefly, mice were anesthetized with 2% isoflurane, endotracheally intubated with a 20G catheter, and ventilated. The transverse aortic arch was surgically accessed. Then, a 25G needle was placed on the transverse aorta, which was secured. Finally, the chest was closed after removing the needle, leaving a stenosis. Mice were administrated with BRL37344 (Tocris Bioscience, Ellisville, Missouri) or SR59230A, respectively, at 0.1 mg/kg/hour via osmotic minipumps.
