Quantification of Opiate Alkaloids in Poppy Seeds

Representative images of poppy seeds and the chemical structures of five OAs are shown in Figure 1 and Figure 2. Morphine, codeine, thebaine, noscapine, papaverine, Morphine-d3, and codeine-d3 were purchased from Sigma (St. Louis, MO, USA). The reference compounds were more than 99% pure, with identity and purity confirmed by chromatographic and spectral data (HRMS). HPLC-grade solvents such as acetonitrile, methanol, isopropanol, and formic acid were procured from Fisher Scientific, Waltham, MA, USA. Ultrapure water (18.2 MΩ.cm) was purified using the Milli-Q system (Millipore, Bedford, MA, USA).

Quantification of Opioids in Urine

Morphine (M-005), Morphine-D3 (M-006), Morphine-D6 (M-086), Codeine (C-006), Codeine-D3 (C-007) and Codeine-D6 (C-041) of 1 mg/ml methanol solution with 99% purity were obtained from Cerilliant (Sigma-Aldrich). Reagents used for the derivation of the analytes and the ISs, Bis (trimethylsilyl) trifluoroacetamide (BSTFA) with 10 g/L trimethylchlorosilane was purchased from Supelco (Bellefonte, PA, USA). Urine samples positive for illicit drugs and used for study analyses were obtained from the Division of Clinical Toxicology, Tri-Service General Hospital. Drug-free urine samples were provided by a member of the research group. All samples were kept at -20°C until analyzed.

HPLC Analysis of Opioid Compounds

Methanol (HPLC grade) was purchased from Merck (Darmstadt, Germany). Ammonium carbonate and formic acid (both analytical grade) were supplied by Acros Organics (Morris Plains, NJ, USA). Acetic acid and ammonium acetate (both analytical grade) were obtained from VWR International (Leuven, Belgium).
Norcodeine, morphine, M3G, morphine-d 3 , codeine-d 3 and M3G-d 3 were obtained from Sigma-Aldrich (St. Louis, MO, USA). Codeine was purchased from Norwegian Medical Depot (Oslo, Norway), and C6G from Lipomed (Arlesheim, Switzerland). M6G was supplied by Chiron (Trondheim, Norway).

Comprehensive PPCP Quantification Protocol

All solvents used in this study were purchased from Fisher Scientific™ (Loughborough, UK) and were of UPLC grade. Individual standards of 30 PPCPs (Table 1), in addition to isotope-labelled Caffeine-D9, Codeine-D3, Carbamazapine-D10, Estone-D4 and 4-Chlorophenol-2,3,5,6-D4 used as internal (surrogate) standards were purchased from Sigma-Aldrich™ (Irvine, UK) at the highest possible purity (>99 %). 13 Ctetrabromobisphenol A ( 13 C-TBBPA) and tris (2-chloroethyl) phosphate-D12 (TCEP-D12) used as recovery (syringe standards) were obtained from Wellington Laboratories (Guelph, ON, Canada). All standard stock solutions were prepared and further diluted in methanol. Oasis MCX and Oasis HLB cartridges (6 cm 3 , 150 mg sorbent per cartridge) were obtained from Waters™ (Hertfordshire, UK). Ammonium formate (NH4COOH), Na2EDTA, ammonium hydroxide (NH4OH, 30 %), ammonium fluoride (NH4F), Acetic acid and formic acid (HCOOH) were obtained from Sigma-Aldrich™ (Gillingham, UK).
Nitrogen gas (oxygen free, 99.998%) was purchased from BOC (Birmingham, UK). Milli-Q water (Merck Millipore, Burlington, MA) was used for cleaning and sample preparation purposes.
