Phase-contrast microscopy of CatIB formation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Phase-contrast microscopic analysis was performed for E. coli BL21(DE3) strains with CatIB formation. 1 µL of each sample was transferred to a microscope slide and covered with a coverslip. The microscope slide was positioned upside down on the desk of an inverted Nikon Eclipse Ti2 microscope (Nikon GmbH, Düsseldorf, Germany). The sample was observed with a CFI Plan Apo Lambda 100X Oil objective (Nikon GmbH, Düsseldorf, Germany) and images were taken with a Thorlab camera DCC154M-GL (Thorlabs Inc., Newton, NJ, US). To determine the percentage of CatIB-producing cells during the cultivation process, cells with visible inclusion bodies at the poles were manually counted (ncells = 45–200).

Quantitative Phase-Contrast Microscopy of E. coli Inclusion Bodies

Check if the same lab product or an alternative is used in the 5 most similar protocols
Phase-contrast microscopic analysis was performed for E. coli BL21(DE3) strains with CatIB formation and for control strains. Cell suspension samples from cultivation experiments were taken before CatIB purification and analyzed by microscopy. A volume of 1 µL was applied on a microscope slide and covered with a coverslip. The microscope slide was positioned upside down on the desk of an inverted Nikon Eclipse Ti2 microscope (Nikon GmbH, Düsseldorf, Germany). The sample was observed with a CFI Plan Apo Lambda 100X Oil objective (Nikon GmbH, Düsseldorf, Germany) and images were taken with a Thorlab camera DCC154M-GL (Thorlabs Inc., Newton, New Jersey, USA). Analysis of cell images were performed with Fiji ImageJ [44 (link)] to determine the areas of at least 100 inclusion bodies and cells.

Microscopic Analysis of CatIB Formation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Phase-contrast microscopic analysis was performed for E. coli BL21(DE3) strains with CatIB formation strains. 1 µL of each sample was given on a microscope slide and covered with a coverslip. The amount of cells that carried CatIBs was counted by analyzing at least 50 cells per CatIB variant.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
