UV-Curable Coatings with POSS Molecules

Coating solutions made from the blend of MA0736 POSS™ molecules (available from Hybrid Plastics Inc., Hattiesburg, Miss.), CN991 (aliphatic urethane acrylate from Sartomer, Exton, Pa.), CN2303 (alkoxylated multifunctional acrylate oligomer from Sartomer), tetrahyhydrofurfuryl alcohol (TFHA) and Lucirin™ TPO-L (photoinitiator, available from BASF) were applied to primed PET film and cured by UV source. The coated film was then treated with oxygen plasma for 90 seconds. The % reflection was measured and reported with the formulations and treatment conditions in Table 4.

Immunoglobulin G Extraction and Proteomic Analysis

Pentaerythritol-tetrakis(3-mercaptopropionate) (“tetrathiol”), triallyl-1,3,5-triazine-2,4,6-(1H,3H,5H)-trione (“triallyl”), 2-(boc amino) ethanethiol, and methanol (99.8%) were obtained from Sigma-Aldrich (Brøndby, Denmark). Lucirin TPO-L was obtained from BASF (Ludvigshafen, Germany). Acetonitrile (ACN), trifluoroacetic acid (TFA), DL-dithiothreitol (D9779, DTT), iodoacetamide (I670-9, IAA), immunoglobulin G (56,834-25MG, IgG, from human serum), trypsin (T1426, from bovine pancreas), and salts were purchased from Sigma-Aldrich (Stockholm, Sweden). L( +)-Ascorbic acid (99.0–100.5%) was from VWR chemicals (Stockholm, Sweden). Water (MQ H2O) was purified in a Millipore Synergy® 185 (Bedford, MA, USA) to a resistivity of 18.2 MΩ·cm at 25 °C. DEAE Affi-Gel® Blue Gel (DEAE) used for IgG extraction was obtained from Bio-RAD, Hercules, USA. Pierce® C18 Tips, 100 µL bed size, were from Thermo Fisher Scientific (Rockford, USA). Kinesis Tubing PEEK (TM) natural 1/32 inch × 0.015 inch connecting syringe and microchip was purchased from Fisher Scientific (Stockholm, Sweden). MALDI matrices 2,5-dihydroxybenzoic acid (DHB) and α-cyano-4-hydroxycinnamic acid (HCCA) and MALDI plate were obtained from Bruker Daltonics (Bremen, Germany).

Synthesis and Characterization of Polymeric Biomaterials

Stannous octoate (tin 2-ethylhexanoate, SnOct 2 ), trimethylolpropane (TMP), trimethylamine, methacrylic anhydride, hydroquinone, methanol absolute, anhydrous dichloromethane and propylene carbonate were purchased from Sigma-Aldrich, Germany. Reagents and solvents were of analytical grade and used as received.

OSTE Devices Surface Modification

The fabricated OSTE devices prepared for capillary filling experiments was rendered hydrophilic by UV grafting of HEMA. A solution of 5% hydroxyethylmethacrylate (HEMA) (Sigma-Aldrich, USA), 0.1% Lucirin TPO-L (BASF Corp., USA), and 0.1% benzophenone (BP) (Sigma-Aldrich, USA) in isopropyl alcohol (IPA) (SunChem AB, Sweden) was prepared. A glass slide was placed above the device, with 150 μm spacing between the top surface of the interlocked pillar array and the glass slide, where the solution was injected. Collimated UV-light was illuminated onto the solution and the pillar array through the glass slide for 300 seconds. After the exposure, the pillar array was rinsed with pure IPA, soaked and incubated with pure IPA for 300 seconds, and dried at room temperature.

Polymer Fabrication for Cell Culture

Methyl methacrylate (MMA), Butyl methacrylate (BMA), Poly(ethylene glycol) dimethacrylate (PEGDA Mw = 550) were obtained from Sigma–Aldrich and used without further purification. Penicilin and streptomycin was provided by EuroClone S.p.a., Italy. C2C12 muscle myoblast cell line was supplied from ATCC® leading biological standards. Human osteoblasts were extracted from human tibia gently donated from Villa Betania Hospital.
