Water, methanol, acetonitrile and 2-propanol were obtained from Carlo Erba (Milano, Italia) while sodium carbonate, sodium bicarbonate and ammonium formate were purchased from Sigma-Aldrich (Milwaukee, WI, USA).

Biochemical Characterization of Sulfotransferase Assay

APAP, adenosine 5’-triphosphate (ATP), dimethyl sulfoxide (DMSO), dithiothreitol (DTT), and N-2-hydroxylpiperazine-N’-2-ethanesulfonic acid (HEPES) were products of Sigma Chemical Company. Morphine, tapentadol, and O-DMT were from Cayman Chemical. Cellulose thin-layer chromatography (TLC) plates were from Merck (EMD Millipore Corporation). Carrier-free sodium [35S]sulfate was from American Radiolabeled Chemicals. 3’-Phosphoadenosine-5’-phospho[35S]sulfate (PAP[35S]) was synthesized using ATP and carrier-free [35S]sulfate according to a previously established protocol [27 (link)]. X-Ray films were from Research Products International Corporation. Prime STAR® GXL DNA Polymerase was a product of Clontech Laboratories, Inc. Protein molecular weight markers were from Bioland Scientific LLC. PCR kit was a product of G-Biosciences. QIAprep® Spin Miniprep Kit was from QIAGEN. Ecolume scintillation cocktail was from MP Biomedical LLC. Glutathione SepharoseTM was a product of GE Healthcare Bio-Sciences. All other chemicals were of the highest grades commercially available.

Receptor Tagging and Ligand Sourcing

The cDNA for the β2 adrenergic, GLP-1, MOR, NOP, and V2 vasopressin receptors, and the cDNA for GRK2, was obtained from the cDNA Resource Center (Bloomsburg University, Bloomsburg, PA). In some experiments receptors modified to include the C-terminal tail of the V2 vasopressin receptor were used. This involved substituting the last C-terminal residues with the last 29 C-terminal residues of the V2 receptor (Oakley et al., 2000 (link)) (last 72 amino acids of the β2 adrenoceptor, and the last 29 amino acids of the MOR, NOP and GLP-1 receptors). DAMGO, met-enkephalin, endomorphin-2, morphine, hydromorphone, oxymorphone, fentanyl, buprenorphine, vasopressin, N/OFQ and exendin-4 were all obtained from Cayman Chemical (Ann Arbor, MI). Isoproterenol was from Millipore Sigma.

Morphine and Naltrexone for Pain Modulation

For in vivo studies, naltrexone was administered intraperitoneally with a dose of 10 mg/kg 2h 30 min before 0.05 mg/mL Morphine solution was injected intrathecally at a volume of 10 μL per mouse 30 min prior to behavior test.
