Renin and Prorenin Measurement in Placental and Cell Lysates

Renin was measured in the placental - and BN16 cell lysates with the Renin III (Cisbio, Gif-sur-Yvette, France) immunoradiometric assay (detection limit 2 pg/mL). Total renin was measured in the lysates with the same assay, after activating prorenin with 10 μmol/L aliskiren (allowing its detection in the assay) at 4 °C for 48 h [45 (link)]. Prorenin in the lysates was calculated by subtracting renin from total renin. Given their low levels, renin and prorenin in the placental perfusates (the latter after its conversion to renin by trypsin) were measured by the more sensitive enzyme-kinetic assay as described before (detection limit 0.05 ng angiotensin I per mL per hour) [46 (link)]. Steroid Profiling and Hormonal Measurements

Plasma concentrations of a panel of 15 steroids were measured using a liquid chromatography coupled to tandem mass spectroscopy (LC-MS/MS) assay to avoid the risk of interference between steroids as previously described [14] . The steroids evaluated were pregnenolone, progesterone, corticosterone, deoxycorticosterone (DOC), aldosterone, 17hydroxypregnenolone, 17-hydroxyprogesterone, 11-deoxycortisol, 21-deoxycortisol, cortisol, cortisone, DHEA, delta 4-androstenedione, 11-β-hydroxyandrostenedione, testosterone.
Plasma renin concentration was assessed with commercial kit (Renin III from CisBio). Urinary concentrations of cortisol and plasma concentrations of ACTH were determined using chemiluminescent enzyme immunoassays (Immulite 2000 XPi, Siemens Healthineers).
Routine plasma and urinary aldosterone concentrations were also assessed with a commercial kit (DPC Coat-a-Count assay from Diagnostic Products Corp).

Hormonal and Renal Function Analysis

Plasma and urine samples were stored at 20 °C until analysis. Plasma renin concentration (PRC) was measured using an immunoradiometric assay (Renin III, CisBio, Codolet, France). Plasma aldosterone concentration (PAC) and 24-h urinary excretion of free aldosterone at pH1 (UAE) were measured using a radioimmunoassay (Coat-a-count, Siemens Medical Solutions Diagnostics, Erlangen, Germany). 24-h urinary free cortisol Excretion (UFCE) was measured by radioimmunoassay (Cortisol RIA kit, Beckman Coulter). The ratio of UAE and UFCE to urinary creatinine excretion was used to standardize the 24hour urine collection. Glomerular filtration rate (GFR) was estimated with the simplified modification of diet in renal disease (MDRD) formula. To account for differences in diet between genotypes and visits, if any, 24-hour urinary sodium and potassium excretion were measured. All biochemical and hormonal assessments were performed blind to the genotype.
