Glucose and Insulin Tolerance Tests in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Glucose tolerance test (GTT) and insulin tolerance test (ITT) were performed in mice before and after a 15-week HFD feeding. For the GTT, mice were fasted for 16 h with free access to water, and then orally gavaged with 1 g/kg body weight of glucose. Blood glucose level was assessed with the Accu-Check Performa (Roche Applied Science, Mannheim, Germany) at 0, 15, 30, 60, 90, and 120 min. For the ITT, mice were fasted for 6 h with free access to water, and then i.p. injected with 1 U/kg of recombinant human insulin (Novolin 30R; Novo Nordisk, Bagsvaerd, Denmark). Blood glucose level was measured at 0, 15, 30, 60, and 120 min after insulin injection.

Mice treated with compounds for 10 days were fasted for 6 h with free access to water. For the glucose tolerance test (GTT), 1 g/kg of glucose was i.p. injected and blood glucose was measured with the Accu-Check Performa (Roche Applied Science, Mannheim, Germany) at 0, 30, 60, 90, 120,150 and 180 min. For the insulin tolerance test (ITT), 1 U/kg of recombinant human insulin (Novolin 30R; Novo Nordisk, Bagsvaerd, Denmark) was i.p. injected, and blood glucose was measured at 0, 30, 60, 120 and 150 min after insulin injection.

Glucose and Insulin Tolerance in Mice

Mice treated with drugs were fasted for 6 h with free access to water. For the glucose tolerance test (GTT), 1 g/kg of glucose was i.p. injected into the mice, and blood glucose was measured with the Accu-Check® Performa (Roche Applied Science, Germany) at 0, 30, 60, 90, and 120 min. For the insulin tolerance test (ITT), 1 units/kg of recombinant human insulin (Novolin 30R, Novo Nordisk, Denmark) was i.p. injected into the mice, and blood glucose was measured at 0, 30, 60, 90, and 120 min after insulin injection. Serum glucose was determined using the Glucose Oxidase Method (APPLYGEN, China), and serum insulin levels were determined by ELISA using an ultra-sensitive mouse insulin kit (Crystal Chem, USA). The cholesterol, Hb1Ac, GSP and ALT levels were assayed using the kits from Nanjing Jiancheng Bioengineering Institute (Nanjing, China).

Tissue Collection and Insulin Response

For collecting mouse tissues, the mice were anesthetized after five weeks of MSC administration, and their epididymal white adipose tissue (EWAT) and liver were isolated and weighed. A portion of mouse livers was promptly cut into small pieces and snap-frozen in liquid nitrogen for below gene expression analyses, while the other part was fixed in formalin for below histological experiments. For western blotting assay, mice were firstly starved for 6 hours, and then administrated with human insulin (1 unit/kg bodyweight, Novolin 30R, Novo Nordisk). One hour later, mouse livers were collected and kept in the liquid nitrogen by snap-freezing for below protein purification.

Glucose and Insulin Tolerance Tests

Check if the same lab product or an alternative is used in the 5 most similar protocols
Intraperitoneal glucose tolerance test (IPGTT) and intraperitoneal insulin tolerance test (IPITT) were undertaken in MSCs- and Saline-injected mice after two weeks of treatment. For the IPGTT, mice were starved for overnight, and their tails were bled to determine the initial blood glucose concentration with a Accu-Chek Mobile glucometer (Roche). Glucose (2 g/kg bodyweight) was administrated into the mice via intraperitoneal injection, and blood glucose level was then monitored. For the IPITT, mice were firstly conducted with 6-hour starvation, then injected with 0.5-1.5 unit/kg of human insulin (Novolin 30R, Novo Nordisk), and the blood glucose concentration was measured.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
