Milk powder was obtained from Bio-Rad Laboratories GmbH (Munich, Germany). Acrylamide (Rotiphorese® Gel 30) was obtained from Carl Roth GmbH (Karlsruhe, Germany). Pure standards for LC-MS analysis of AEA, 2-AG, OEA, PEA and AEA-d8 were purchased from Cayman Chemical (Ann Arbor, MI, USA).

Reagent Procurement for Cell Culture Assays

Lovastatin hydroxy acid, sodium salt, was provided from Toronto Research Chemical (Toronto, Canada) and Triton® X-100, acetonitrile (LC-MS grade) and trifluoroacetic acid (analytical grade) from Roth (Karlsruhe, Germany). Penicillin-streptomycin was from Invitrogen (Darmstadt, Germany). Dulbecco's Modified Eagle's medium (DMEM) with 4 mM L-glutamine and 4.5 g/L glucose was from Lonza (Cologne, Germany). Chemiluminescence Assay for ROS Quantification

The ROS levels in culture media were evaluated by chemiluminescence assay (26 (link)) using 5-amino-2,3-dihydro-1,4- phthalazinedione (luminal; Sigma, St. Louis, MO, USA) as a probe in a luminometer (Synergy H1 Hybrid Reader; Biotek, USA). Each sample was scanned at 37°C for 15 minutes by the luminometer. ROS values were expressed as relative light units (RLUs). All the samples were measured in duplicate.

Measuring Chitin-Induced ROS Burst in Rice

The method for measuring the chitin-induced ROS burst levels is described in [54 (link)]. Briefly, leaf discs were cut from 7-day-old rice seedlings and floated on water overnight, and then the water was removed. After that, the leaf discs were incubated with 100 µL reaction solution (20 µM luminal, Sigma Aldrich, Saint Louis, MO, USA, Cat. No. 123072 and 2.5 µg/mL peroxidase, Solarbio Science & Technology Co., Ltd. Beijing, China, Cat. No. P8020) containing 400 nM chitin (Sigma-Aldrich, Saint Louis, MO, USA, Cat. No. H1396) to immediately detect the ROS burst. The luminescence was measured by a Berthold Mithras luminometer (Berthold Technologies GmbH & Co.KG, Bad Wildbad, Germany) every 1~2 min for 55 min. Eight replicates were performed for each sample.

Luminescent Measurement of H2O2 in Tomato Leaves

The peptides AtPep1 and flg22 with purity of over 95% were synthesized by China Peptides Company Limited. Tomato leaf disks of 3 mm at diameter were dipped in 200 μl of distilled water in a 96-well plate in dark over night. Water was replaced with 200 μl solution containing 100 μM luminal (Sigma-Aldrich) and 1 μg of horseradish peroxidase. AtPep1 (10 nM) and flg22 (100 nM) were added and immediately H2O2 were measured for 30 min as luminescence using a luminometer (Glomax 96 microplate luminometer, Promega).
