Remdesivir, GS-441524, and its heavy isotope internal standards were purchased from Alsachim (Illkirch-Graffenstaden, France). Lactose monohydrate, leucine, polysorbate 20, acetonitrile (HPLC grade), and trifluoracetic acid were purchased from Fisher Scientific (Pittsburgh, PA, USA). D-Mannitol was bought from Acros Organic (Fair lawn, NJ, USA). Dipalmitoylphosphotidylcholine (DPPC) was purchased from Avanti Polar Lipid, Inc. (Alabaster, AL, USA). Sulfobutylether-beta-cyclodextrin (SBECD, Captisol®) was kindly provided by CyDex Pharmaceuticals, Inc. (Lawrence, KS, USA) High-resistance Monodose RS00 dry powder inhalers were kindly provided by Plastiape S.p.A. (Osnago, Italy).

Aggregation Susceptibility of Monoclonal Antibody

Humanized IgG1 monoclonal antibody (molecular weight 146 kDa), here denoted as “3M”, was provided by MedImmune (Gaithersburg, MD).37 (link) The antibody was obtained at a stock concentration of 150 mg/mL in 10 mM L-histidine at pH 6. The antibody 3M is a human IgG1 with three mutations (S239D/A330L/I332E) in the CH2 portion of the Fc. These mutations reduce the thermal stability of 3M,37 (link) which was chosen for the current work because of previous studies36 (link) that showed it to be prone to aggregation when exposed to silicone oil-water interfaces. Polysorbate 20 (>97% purity, Fisher BioReagents) was obtained from Fisher Scientific (Pittsburgh, PA). All buffer salts were of reagent grade or higher, and all solutions were prepared in de-ionized water filtered through a 0.22 μm Millipore filter (Billerica, MA). Silicone oil (Dow Corning 360, 100 cSt) was of medical grade and purchased from Nexeo Solutions (Denver, CO). The syringes used in the incubation studies were BD Hypak SCF 1mL long 27G1/2 (BD Medical-Pharmaceutical Systems, Franklin Lakes, NJ). Glass coverslips, Micro-90, and isopropyl alcohol were obtained from Fisher Scientific (Waltham, MA). Nickel TEM grids (EMS G100-Ni) were obtained from Electron Microscopy Sciences (Hatfield, PA). Teflon® rings were fabricated in-house at the University of Colorado-Boulder.

The following materials were purchased from Sigma Aldrich (St. Louis, MO): 10x phosphate buffered saline (PBS), DL-dithiothreitol (DTT), sodium dodecyl sulfate (SDS), sodium sulfate, sodium phosphate dibasic, and acrylamide. Materials from Thermo Fisher Scientific (Waltham, MA) included polysorbate 20, tris(hydroxymethyl)aminomethane, glycine, and sodium phosphate monobasic.
