mRNA and miRNA Expression Profiling

Hybridizations of mRNAs and miRNAs were performed as previously reported by our group [22 (link)–24 (link)]. The oligo microarrays technology used for gene expression assays was the SurePrint G3 Human Gene Expression v3 8x60K oligo microarrays (G4851C, Agilent Technologies) and the Human miRNA Microarray Kit (V3) (8x15K-G4471A, Agilent Technologies) was used to assess the expression of miRNAs. For both microarrays, the total RNA amount (200 ng) and the one color (cyanine-3, Cy3) Quick Amp labeling kit (Agilent) was used. The hybridization steps varied according the manufacturer’ instructions, with 17 hours at 65C for mRNA and 24 hours at 55C for miRNA microarrays. Images were acquired using an Agilent DNA microarray scanner with SureScan technology (Agilent Technologies).
