Acetylcholinesterase (AChE) from Electrophorus electricus, acetylthiocholine iodide, 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), aluminum nitrate, butyrylcholinesterase (BChE) from equine serum, S-butyrylthiocholine chloride, 3-O-caffeoylquinic acid (chlorogenic acid), catechin, 2,2-diphenyl-1-picrylhydrazyl radical (DPPH), 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB), ellagic acid, Folin-Ciocalteu phenol reagent, ferric chloride, galantamine hydrobromide, gallic acid, 4-hydroxybenzyl alcohol, 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox), myricetin, 2,4,6-tris(2-pyridyl)-1,3,5-triazine (TPTZ), and quercetin were purchased from Sigma Aldrich (Milan, Italy). LC-MS grade acetonitrile and formic acid were purchased from Merck (Darmstadt, Germany). Standards of myricetin-3-O-glucoside, quercetin-3-O-glucoside, and acacetin were purchased from Extrasynthese (Genay, France). HPLC grade water (18 MΩ·cm) was prepared by using a Millipore (Bedford, MA, USA) Milli-Q purification system.
Spectrophotometric determinations were obtained with an Ultrospec 2100 spectrophotometer (Biochrom Ltd., Cambridge, England) using cells with a 1 cm path length.

Standardized Phytochemical Characterization

The standard compounds and reagents (all of analytical grade): MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), DMSO (dimethyl sulfoxide), gallic acid, Na2CO3, 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), Folin-Ciocalteu’s reagent, resazurin, tetracycline, chloramphenicol, kanamycin, acetonitrile (LC-MS grade, gradient grade), formic acid, 4-methoxybenzoic acid, 4-hydroxy-3-methoxycinnamaldehyde, caffeic acid, quercetin, luteolin and pinostrobin PBS were purchased from Merck (Darmstadt, Germany). The standards of 4-hydroxybenzoic acid, p-coumaric acid, ferulic, isoferulic acid, pinobanksin, chrysin, sakuranetin, naringenin, apigenin, kaempferol, isorhamnetin, acacetin and pinocembrin were obtained from Extrasynthese (Genay, France) and galangin from Alfa Aesar, (Haverhill, MA, USA). The antibiotics: ampicillin, oxacillin, teicoplanin, gentamicin, amikacin, fusidic acid, erythromycin, mupirocin, rifampicin, levofloxacin, norfloxacin, linezolid, bacitracin were purchased from Argenda (Poland). Methanol, ethanol, vanillin, benzoic acid, cinnamic acid were purchased from (POCH, Gliwice, Poland). Ultrapure H2O (18.0 MΩ) was obtained with a Milli-Q Advantage A10 system (Millipore, Billerica, MA, USA). The absorbance of the reaction mixture in Folin-Ciocalteu, DPPH assays were measured using a Genesys 20 spectrophotometer (Thermo Scientific, Waltham, MA, USA).

Acetonitrile, water, and formic acids (all of LC-MS grade) were purchased from Merck (Darmstadt, Germany).

Characterization of Plant-Derived Compounds

Practically all solvents, salts, mediums, and reagents were from Sigma-Aldrich (Milan, Italy). In addition benzophenone-3, rutin, quercetin, trans-ferulic and chlorogenic acids, and silibinin were from Sigma-Aldrich. Acacetin, baicalein, chalcone, cyanidin chloride, chrysin, and (+)-taxifolin were purchased from Extrasynthese Co. (Lyon, France) and resveratrol from Biomol (Research Lab, Plymouth, MA). Verbascoside (>97% purity) was isolated from meristem plant cells of Syringa vulgaris, and leontopodic acids (>98% purity) were from meristem plant cells of Leontopodium alpinum (IRB S.r.l., Altavilla Vicentina, Italy). Other reagents, primers, and antibodies are mentioned herein below in the appropriate subsections.

Modulation of IL-2 Production in Activated T Cells

In total, 1 × 105 clone 1C116 or 2C21 cells, were cultured with each material (concentration ranges are described below) or vehicle control (dimethyl sulfoxide: DMSO) and 20 ng/mL PMA in 96-well U-bottom tissue culture plates in 200 μL of CCM for 24 h. The materials were 0–100 μg/mL flavonoids (quercetin, isoquercitrin, hyperoside, galangin, kaempferol, morin, myricetin, hesperetin, hesperidin, acacetin, linarin, isorhoifolin, (all purchased from Extrasynthese, Lyon, France)), 0–1000 μM IPP (Sigma-Aldrich), 0–2000 μM risedronate, 0–33.5 μM methylamine (Sigma-Aldrich), 0–33.5 μM ethylamine (Sigma-Aldrich) and 1 μg/mL purified anti-human CD3 mAb (OKT3: Thermo Fisher Scientific). After the incubation, the culture supernatant was corrected, and the concentration of IL-2 was analyzed using an ELISA kit (BD Biosciense).

(St. Louis, MO, USA). Tyrosol, protocatechuic acid, sinapic acid, o-coumaric acid, and caffeic acid, and epicatechin were purchased from Fluka (Steinheim, Germany); vanillic acid was obtained from Serva (Heidelberg, Germany), while kaempferol, chrysin, naringenin, acacetin, and apigenin was obtained from Extrasynthese (Genay-Cedex, France). Ethanol/water mixture used for the extraction of sour cherry (Prunus cerasus L.) was a distillation product from local producers.
