TUNEL staining was conducted using the DEADEnd TUNEL staining kit (Promega Cat# G3250). Sections were deparaffinized in a graded ethanol series, re-fixed in 10% Formalin, and permeabilized with 20 ug/ml of Proteinase K. All other steps were followed according to the manufacturer’s instructions. Sections were counterstained with DAPI (Vector Laboratories, Inc, Burlingame, CA Cat # H-1200). Mesenchymal and ectodermal TUNEL positive cells were quantified independently.
