96 well microplates were coated with 0.5 μg/well of LEAF™ Purified anti-mouse CD3ε (BioLegend) and incubated for 2h at 37°C. Wells were washed to remove unbound antibody. C57BL/6J splenocytes were sorted by flow cytometry as described above and CD3+ T-cells were negatively selected. 2×105 cells were added to each well and incubated at 37°C for 30 min. Following incubation, 0.4 μg soluble LEAF™ Purified anti-mouse CD28 (BioLegend) was added to each well for a final concentration of 2 μg/mL. Cultures were incubated for 48 hr at 37°C, pulsed with 1μCi/well [3H]-thymidine, and harvested 18 hr later, and radioactive uptake measured by liquid scintillation counting using a Packard 1900 TR liquid scintillation counter.

Isolation and Activation of Murine CD4+ T Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Splenic mononuclear cells were isolated from mice by mouse lymphocyte separation medium (Dakewei, Shenzhen, China) as previously described (17 (link)). Single-cell suspensions were separated by mechanical disruption of mouse spleens through 70-μm cell strainers and were harvested by gradient centrifugation at 800 g for 30 min, and washed in PBS twice for the experiment. Purified CD4+ T cells were negatively selected using the MojosortTM Mouse CD4+ T Cell Isolation kit (Biolegend, San Diego, CA, USA). The purity of CD4+ T cell was > 95% (Supplementary Figure S1A), and the cells were cultured in RMPI 1640 medium with recombinant murine IL-2 (20 IU/ml, Novoprotein, Shanghai, China). The cells were stimulated with plate-bound anti-CD3 (2.5 µg/ml, LEAF™ Purified anti-mouse CD3ϵ, Biolegend, San Diego, USA) and soluble anti-CD28 (2.5 µg/ml, LEAF™ Purified anti-mouse CD28, Biolegend, San Diego, USA), and the cells were incubated for 24 h for further experiments.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
