Cell Migration and Invasion Assay

Cells were harvested using 0.25% trypsin-EDTA. The solution was then centrifuged and cells were resuspended in a serum-free media containing 0.1% BSA. Six hundred microliters of complete medium containing 10% FBS was then added to the lower layer of Transwell 24-well plates (Millipore, Burlington, MA, USA). Cell suspension in the upper layer (200 µL, 4×105 cells) was incubated at 37°C and 5% CO2. Migration and invasion were assessed after 24 and 50 hours, respectively, by fixing the cells with methanol for 30 minutes and staining the cells with 0.1% crystal violet for 15 minutes.

Migration (without Matrigel) and invasion (with Matrigel) assays were conducted using transwell 24-well plates (8-μm pores; Millipore). A total of 5 × 10 4 cells were suspended in the upper chamber in 150 μL serum-free medium, and 600 μL normal growth medium was placed in the lower chamber. Cells which migrated/invaded through to the under-side of the membrane were xed, stained, and counted in 5 random elds of view.
