Electroretinography in Dark-Adapted Mice

Mice were dark-adapted before electroretinography (ERG). For general anesthesia, the animal was injected intraperitoneally with a mixture of Ketamine and Xylazine (100 and 10 mg/kg body weight, respectively) in PBS. The pupil was locally anesthetized with 0.5% proparacaine hydrochloride (Alcon, Belgium) and dilated with 1% Mydriacyl (Alcon, Belgium). White flash of 3 (p)cd.s/m2 and 10 (p)cd.s/m2 intensities were delivered from the ColorDome Ganzfeld System (Diagnosys, United States). a-wave, b-wave, and the ascending phase of the b-wave (oscillatory potentials, OPs) were recorded using the Espion ERG Diagnosys and analyzed by Epsion V5 System (Diagnosys software) as previously reported (Li et al., 2012 (link); Yang et al., 2017 (link); Wang et al., 2020 (link)).

Phentolamine Mesylate for Dry Eye Relief

Subjects were randomized in a 2:1 ratio to the phentolamine mesylate and placebo treatment group, respectively. Patients in the placebo group received one drop of polyquad lubricant eye drop (Tears Naturale IIⓇ, Alcon, Fort Worth, TX) in each eye. Patients in the treatment group received one drop of 1.0% phentolamine mesylate prepared in Tears Naturale II® vehicle (PMOS) in each eye. Scotopic Electroretinogram Measurement Protocol

Electroretinogram (ERG) was performed as previously described.13 (link) Animals were dark-adapted overnight and then anesthetized under dim red light with the ketamine/xylazine as described above. One per cent (wt/vol.) Tropicamide (Mydriacyl, Alcon-Couvreur, Puurs, Belgium) was applied topically to dilate the pupils. Moisturizing eye drops (Tears Naturale II, Alcon-Couvreur, Puurs, Belgium) were used to keep the cornea lubricated. Topical anesthetic proparacaine hydrochloride (0.5% (wt/vol.), Alcaine, Alcon-Couvreur, Puurs, Belgium) was applied to the cornea as needed. For ERG signal recording, gold ring electrodes were placed on the cornea. Subcutaneous needle electrodes were inserted at the dorsal midline between two ears and the tail serving as the negative and ground references, respectively. Scotopic ERG responses were recorded using the Color Dome Ganzfeld System (Diagnosys Espion system, Diagnosys, Lowell, Massachusetts, USA) at light intensities of 3.0 and 10.0 cd s/m2, respectively. Amplitudes of a-wave and b-wave were analyzed.

Topical Antibiotic and Steroid Therapy for Ocular Healing
