Culturing Human Skin Cells

Human epidermal keratinocytes and human dermal fibroblasts were obtained from Lonza (Walkersville, MD). Human epidermal keratinocytes were cultured and expanded in serum-free keratinocyte growth medium (Lonza), and human dermal fibroblasts were cultured and expanded in Dulbecco’s modified Eagle’s medium (DMEM, Gibco, NY) with 10% (v/v) fetal bovine serum (FBS, Gibco BRL) and 1% (v/v) penicillin/streptomycin (Sigma-Aldrich, St. Louis, MO). Cells were cultured at 37°C in a humidified 5% CO2 incubator. The medium was changed every 2–3 days until the use.

Human Skin Cell Culture Protocol

Human adult hDFs and human epidermal keratinocytes were purchased from Lonza Walkersville Inc (Walkersville, MD, USA). hDFs were cultured in Dulbecco's Modified Eagle's Medium (DMEM) containing 1% penicillin-streptomycin and 10% fetal bovine serum (FBS) (Sigma-Aldrich, St. Louis, MO, USA). For assays, hDFs at 3 passages were seeded on 12- or 48-well plates (BD, Franklin Lakes, NJ). human epidermal keratinocytes were cultured in KGM-Gold™ Keratinocyte Growth Medium SingleQuots™ Supplements and Growth Factors (Lonza, Basel, Switzerland). At 2 passages, these keratinocytes were seeded on 12- or 48-well plates (BD) for use in assays. Interleukin (IL)-1β (1 ng/ml) (AF-200-01B, PeproTech, Rocky Hill, NJ) was added 24 h after passage.
