Comparison of CMV PCR Extraction Methods

Check if the same lab product or an alternative is used in the 5 most similar protocols
For the reference method, 100 µl of each specimen was extracted on the MagNA Pure LC (Roche, Indianapolis, IN) using the MagNA Pure LC DNA isolation kit with the DNA I Blood Cell High Performance protocol. The CACM assay was set-up manually using 50 µl out of the 100 µl elution.
For the test methods, 400 µl of specimen was extracted on the QIAsymphony SP (Qiagen, Germantown, MD) using the Qiagen Virus/Bacteria Midi kit and Pathogen Complex 400 protocol. Specimens were also extracted using the same kit and the Cell-free 1000 protocol, which extracts from 1000 µl of sample. For eight negative tissue specimens, there was insufficient volume for extraction using the Cell-free 1000 protocol. The artus CMV PCR assay (Qiagen, Germantown, MD) was set-up with a custom protocol on the QIAsymphony AS that utilizes 10 µl of the 95 µl elution, 12.5 µl CMV RG Master, and 2.5 µl of CMV Mg-Sol, for a final reaction volume of 25 µl.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
